This dissertation is mainly composed of three chapters.In the first chapter,the chemical constituents of Elsholtzia bodinieri,belong to family Labiatae,were systematically studied.All the identified compounds were investigated for their anti-inflammatory activity in LPSinduced RAW 264.7 cells.In the second chapter,the total alkaloids of G.lanceolata,a plant of Gardneria belong to family Loganiaceae,were isolated and identified.In the third chapter,the chemical constituents and biological activities of the genus Gardneria were reviewed.By using various chromatographic techniques(including normal and reverse phase silica gel,TLC,medium pressure liquid,MPLC、HPLCSephadex LH-20),and diverse spectroscopic methods including nuclear magnetic resonance,mass etc,38 non-repeating compounds were isolated and identified from the above two plants,including triterpenoids,flavonoids,sesquiterpenoids,lignin,steroids,monoterpene indole alkaloids,etc.Some compounds had significant anti-inflammatory activities,further clarified the material basis of E.bodinieri and G.lanceolata,providing theoretical and experimental basis for future development.Chapter Ⅰ Chemical constituents from Elsholtzia bodinieri and their anti-inflammatory activitiesElsholtzia bodinieri,belong to family Labiatae,is mainly distributed in Yunnan,which is used as medicine and food.It is conventionally served as folk medicines to treat exogenous cold,deficiency fire toothache,indigestion and so on.There are few chemical studies on Elsholtzia bodinieri at home and abroad.Therefore,systematic chemical studies had been carried out on the whole plant of Elsholtzia bodinieri,and 28 non-repeating monomer compounds were isolated from the ethyl acetate layer of Elsholtzia bodinieri,the antiinflammatory activity of all isolated compounds was evaluated in LPS-induced RAW 264.7cells.As a result,most of the compounds had certain anti-inflammatory effects.At the concentration of 20 μg/m L,compounds 1~5,7,9~13,15,16,19,21~27 have obvious inhibitory effects on the release of NO.Compared with the positive control dexamethasone,compounds 2 and 12 have better inhibitory effects on the release of NO than that of dexamethasone.According to the inhibition of NO release,compounds 1,2,5,9,10,15,16,19,22~27 were further evaluated for their inhibitory effect on the production of IL-6,TNF-α and IL-1β.Compounds 1,5,9,10,15,19,23,24,26 showed significantly inhibitory effect on TNF-α production,especially compounds 1,10,19 and 24 displayed more inhibitory effect than that of the positive control group;Compounds 1,2,5,9,10,15,16,19,22~27 showed significantly inhibitory effect on IL-6 production,and compounds 2,5,15,23~26 showed higher inhibition rate than that of the positive control group;Compounds 1,2,5,9,10,15,19,22~26 presented markedly inhibitory effect on IL-1β production,while compounds 15 and 23 showed a better inhibition rate,Compounds 1,5,9,10,15,19,23,24,26 have good inhibitory effects on these three inflammatory factors.Chapter Ⅱ Chemical constituents of Gardneria.lanceolataGardneria lanceolata is traditionally applied as herb for treatment diversity ailments,and belongs to the family Loganiaceae.It is widely used in folk medicine to treat the pain of waist and knee,rheumatic arthralgia,kidney deficiency,enuresis,scrotal dampness and other diseases.In order to further reveal the chemical composition of G.lanceolata,it is expected to find more novel monomer compounds.In this experiment,10 compounds were isolated from the total alkaloids of G.lanceolata.The structural types of the compounds included 9monoterpene indole alkaloids and 1 phytosterol,of which compound 1 was a new compound.Chapter Ⅲ Research progress in chemical constituents and pharmacological activities of GardneriaThis chapter summarizes the chemical components and bioactive components of the plants of the genus Gardneria from 1967 to present,providing a theoretical basis for the future development of the genus.
