| Platycodon grandiflorus is a kind of medicinal and food material,which is favored in daily life for its unique flavor and good pharmacological effects.As a relatively common Chinese medicine,the pharmacological effects of Platycodon are mainly to promote lung,expectorant,pharyngeal,and pus drainage,and are used to treat cough with phlegm,chest congestion,sore throat,mute voice,and canker sores and pus.The main active ingredients of Platycodon grandiflorus are saponins,and it also contains small amounts of flavonoids,polysaccharides and anthocyanins.In order to fu Lly exploit the plant resources of Platycodon grandiflorus,the separation,extraction and efficacy of the functional components of Platycodon grandiflorus have attracted much attention.In this paper,we conducted a systematic study on the extraction technique of Platycodon grandiflorus saponin and investigated the alleviating effect and mechanism of Platycodon grandiflorus saponin in mice with gouty arthritis.(1)The process and effect of solvent extraction,u Ltrasonic extraction,and microwave-assisted extraction of Platycodon grandiflorus saponin were studied,and the best effect of microwave-assisted extraction of Platycodon grandiflorus saponin was determined.The process conditions and parameters of microwave extraction of saponin were optimized on the basis of single-factor experiments,and response surface experiments were conducted to determine the better extraction conditions for microwave power 210 W,extraction time 3.5 min,solid-liquid ratio of 1:17.5,and ethanol concentration of 80%,in which the total saponin extraction rate of 5.47%(for the dried base of Platycodon grandiflorus saponin),saponin The saponin content was76.2%.(2)The purification of Platycodon grandiflorus saponin using D101 resin,the ion exchange resin static adsorption time,sample concentration,sample volume,sample flow rate,as well as the pre-elution rinse water,elution flow rate,elution volume,eluent concentration and other technical indicators were optimized to determine the resin purification conditions: The static adsorption time was 5 h,the upper sample volume was 50 m L,the upper sample raw drug concentration was 0.02g/m L,the upper sample flow rate was 1.4 m L/min,the rinse water volume before elution was 330 m L,the elution flow rate was 3.4 m L/min,the concentration of the eluent was 80% ethanol,and the elution volume was 210 m L.the saponin content in the sample was increased from about 76.2% to the purified 91.3%.(3)The effect of Platycodon saponin on mice with gouty arthritis was studied.The mice were acclimatized and fed for 7 days and grouped into blank group,model group,positive control group and low,medium and high concentrations of Platycodon grandiflorus saponin.The blank and model groups were gavaged with saline,and the positive control group was gavaged with colchicine for 7 days,and the gouty arthritis model was built by MSU injection on day 5,and the effects of Platycodon grandiflorus saponin on gouty arthritis mice were investigated.The resu Lts showed that origanum saponin cou Ld significantly improve the gait index and the morphology of damaged cells at the ankle,and was able to reduce the degree of joint swelling and the level of myeloperoxidase in mice with gouty arthritis.(4)Studies have analyzed the mechanism of gout arthritis alleviation by Platycodon grandiflorus saponin.The results showed that Platycodon grandiflorus saponin can effectively reduce the level of inflammatory factors,inhibit the expression of related proteins,and reduce the level of oxidative stress,through these studies,it can be demonstrated that Platycodon grandiflorus saponin can inhibit the expression of related proteins on the NLRP3/ASC/caspase-1 signaling pathway,thereby inhibiting the inflammatory response,and thus play a role in the relief of gouty arthritis,and its mechanism of action is consistent with the existing gout treatment main The mechanism of action is consistent with that of colchicine,the main drug for gout treatment. |
