Bioinformatics Analysis Of Differentially Expressed Genes And Related CircRNA On Cutaneous Squamous Cell Carcinoma

Abstract/Summary:
Objective: To analyze the expression of pathogenic genes and related circRNA in cutaneous squamous cell carcinoma by bioinformatics technology,explore the mechanism of occurrence and development of cutaneous squamous cell carcinoma and provide reference ideas for finding new therapeutic targets.Methods: Cutaneous squamous cell carcinoma was searched in GEO database,and the human genes obtained from skin tissue in recent ten years were selected for analysis.After screening and using principal components analysis to remove the batch effect,this paper selects three sets of data sets: GSE108008,GSE191334 and GSE32979 for analysis.GSE108008 data set selected 10 cases of skin exposure and 10 cases of cutaneous squamous cell carcinoma gene data;GSE191334 data set selected 8 cases of cutaneous squamous cell carcinoma gene data with normal skin matching;GSE32979data set included organ transplant recipients’ gene data,including 15 cases of cutaneous squamous cell carcinoma samples and 13 cases of normal samples.The differential genes were screened by differential expression analysis,and the functional enrichment were analyzed by using the statistical hypergeometric distribution analysis Then PPI network were constructed and analyzed.The circRNA-related gene information from human skin tissue in recent ten years,namely GSE74758 data set,was selected from GEO database,which included 3 cutaneous squamous cell carcinoma samples and 3 normal skin samples.After screening the related differential circular RNA by differential expression analysis,the ce RNA network was constructed,and the functional enrichment and PPI network were constructed and analyzed.Pubmed,CNKI,Cochranelibrary,Clinicaltrials and Wanfang database were searched for all the literatures about immunohistochemistry,PCR,cell transfection and animal experiments related to cutaneous squamous cell carcinoma since the establishment of the databases.The literatures containing the differential genes and the related genes predicted by differential circRNA were screened,and then the expressions of genes in cutaneous squamous cell carcinoma tissues and cell lines and in normal skin tissues and cell lines were collected and counted,and the difference was analyzed by chi-square test.Results: Compared with normal skin,375 genes related to cutaneous squamous cell carcinoma were abnormally expressed,including 147 differentially up-regulated genes and 228 differentially down-regulated genes as well as 14 hubgenes were screened,including CXCL8、S100A7、SPRR1B、SPRR3、FLG、CXCL10、CCND1、CSTA、PTGS2、PPARG、CD80、SPP1、IRF1、CXCL5.9 related circRNAs were abnormally expressed,among which 1 up-regulated circRNA,including hsa_circ_0001402 and 8 differentially down-regulated circRNAs including hsa_circ_0022392 、 hsa_circ_0022383 、 hsa_circ_0005085 、 hsa_circ_0001410 、hsa_circ_0089252、hsa_circ_0007928、hsa_circ_0015449 and hsa_circ_0076251.A total of 528 genes were predicted and 10 hubgenes were screened after construction of ce RNA network,including PTEN、KRAS、STAT3、SIRT1、ITGB1、RAB5A、FOS、YWHAZ、CAV1 、 TGFBR1.After the statistics and analysis of 71 articles about cutaneous squamous cell carcinoma,it is concluded that the expression of some of the differential genes screened in this paper and some of the related genes predicted by differential circRNA in this paper in normal skin tissues and cell lines and in cutaneous squamous cell carcinoma tissues and cell lines.This conclusion is basically consistent with the results of this paper.Conclusion: In this paper,bioinformatics analysis was used to find that the occurrence and development of cutaneous squamous cell carcinoma was related to the regulation of many genes and circRNA.14 hubgenes were screened by differentially expressed genes.The top three hubgenes were CXCL8,S100A7 and SPRR1 B.10 hubgenes were screened by predicted circRNA hubgenes,and the top three hubgenes were PTEN,KRAS and STAT3.At present,there is no overlap between the differentially expressed genes in cutaneous squamous cell carcinoma and the genes predicted by differentially expressed circRNA.
