| Objective: Oral squamous cell carcinoma(OSCC)is the most common oral cancer in clinic.Despite the continuous improvement of treatment methods,the rate of mortality has not reduced naturally,as well as the postoperative metastasis and recurrence rate are still high.So,it is necessary to illuminate its pathogenesis at the molecular level and explore new biomarkers with high sensitivity and specificity,so as to detect and diagnose OSCC accurately.Circular RNA is a type of non-coding RNA with the characteristics of wide expression,high conservation and high stability and is considered to be a new molecular biomarker for clinical diagnosis,treatment and prognostic evaluation,and it has great potential in precise targeted therapy.Therefore,this study aims to explore the expression and possible biological functions of hsa_circ_0087960 in the occurrence and development of oral squamous cell carcinoma,and provide new ideas and new targets for the treatment of oral squamous cell carcinoma.Methods: Analyzed the expression profiles of circRNAs and mRNAs in OSCC tissues through high-throughput sequencing.Three online bioinformatics analysis tools were used to explore mostly probable biological functions and pathways of the target genes of hsa_circ_0087960 in oral squamous cell carcinoma.Subsequently,verified the differential expression of hsa_circ_0087960 in OSCC tissues and cells,and further analyzed the correlation between hsa_circ_0087960 and clinicopathological features of OSCC.At last,the ROC curve was created to analyze the diagnostic potential of hsa_circ_0087960 in OSCC and a circRNA-miRNAs-mRNAs network was constructed.Results: Based on bioinformatics analysis results showed three mostly probable functions(cellular process,single-organism process,binding and catalytic activity)and probable pathways(pathways in cancer,PI3K-Akt signaling pathway and platelet activation).Verified by real-time quantitative PCR experiment showed that the hsa_circ_0087960 in OSCC was significantly down-expressed.Combined with the analysis of clinicopathological data,patients with lower expression of hsa_circ_0087960tended to have increased tumor sizes and advanced TNM stage of oral squamous cell carcinoma.By calculating the area under the receiver operating characteristic curve,the expression of hsa_circ_0087960 has the diagnostic efficiency of 67.9% for OSCC cases.Conclusions: 1.Hsa_circ_0087960 is significantly low expressed in oral squamous cell carcinoma tissues and cell lines.2.Hsa_circ_0087960 has certain differential diagnosis value for oral squamous cell carcinoma and non-cancerous tissue.3.Hsa_circ_0087960can be used as a potential biomarker and is expected to become a future therapeutic target for OSCC treatment. |
