Expression And Clinical Significance Of TRIM8 In High-grade Serous Ovarian Carcinoma

Objective:In order to offer an experimental and theoretical foundation for early diagnosis and targeted treatment of high-grade serous ovarian cancer,it is important to investigate the expression of trigonal motif 8(tripartitemotif8,TRIM8)in high-grade serous ovarian cancer.Methods : From January 2018 to December 2021,76 patients who underwent ovariectomy at the first hospital of China Medical University were chosen,and their ovarian pathological tissues were studied.According to the pathological results,76 patients without neoadjuvant chemotherapy were divided into high grade serous ovarian cancer group(n = 35)and benign serous cystadenoma group(n = 20).Forty-one patients with high-grade Figo stage Ⅲ-Ⅳ serous ovarian cancer were divided into neoadjuvant chemotherapy group(n = 21)and non-neoadjuvant chemotherapy group(n = 20).The expression of TRIM8 in the above patients was detected by immunohistochemical method,and the difference of TRIM8 expression between groups and its relationship with clinicopathological features were compared and analyzed.The correlation between the expression of TRIM8 and the prognosis of patients was analyzed by Kaplan-Meier Plotter online database.Results:1.In high grade serous ovarian cancer and ovarian benign serous cystadenoma,the expression frequencies of TRIM8 protein were 62.9%(22/35)and 0.5%(1/20),respectively;2.The findings demonstrated that high grade serous ovarian cancer considerably out-expressed benign serous cystadenoma in terms of TRIM8 protein expression(P<0.05)and sensitivity,specificity and accuracy of TRIM8 protein in high grade serous ovarian cancer were 62.9%,95.0% and 74.5%,respectively.Besides,the Yoden index is 0.58 and the area under the ROC curve is 0.78;3.The analysis of the clinicopathological features of TRIM8 protein and high-grade serous ovarian cancer showed that the expression of TRIM8 protein was related to clinical stage,menopause and neoadjuvant chemotherapy(P<0.05),but not with age,maximum diameter of lesion,serum CA125 level and omental metastasis(P > 0.05);4.The results of Kaplan-Meier Plotter online database analysis showed that the patients with high expression of TRIM8 in serous ovarian cancer had a worse prognosis than those with low expression,which was related to the poor prognosis of serous ovarian cancer(P > 0.05).Conclusions:1.The expression of TRIM8 protein is different between high-grade serous ovarian carcinoma and benign serous cystadenoma,and its specificity is high;2.The positive expression rate of TRIM8 protein in TRIM8 in postmenopausal patients was significantly higher than that in non-menopausal patients.The positive expression rate of TRIM8 protein in patients with FIGO stage Ⅰ-Ⅱ was significantly lower than that in patients with Figo stage Ⅲ-Ⅳ.The positive expression rate of TRIM8 protein in patients with neoadjuvant chemotherapy before operation was significantly lower than that in patients without neoadjuvant chemotherapy;3.The results of Kaplan-Meier Plotter online database analysis showed that the high expression of TRIM8 was associated with poor prognosis in patients with serous ovarian cancer.
