Association Of PPIA Gene Polymorphism At The Rs6850 Locus With Type 2 Diabetes Mellitus Combined With Coronary Artery Disease

Abstract/Summary:
Objective: To study the correlation between single nucleotide polymorphism of PPIA gene rs6850 locus and T2 DM combined with coronary artery disease,which can be used to guide clinical personalized medication and provide new diagnostic ideas and research directions for preventive medicine,clinical medicine and genetic medicine.Methods: A total of 200 patients(50 with stable angina,50 with ACS,50 with T2DM+stable angina,50 with T2DM+ACS)with coronary artery disease and type 2 diabetes combined with coronary artery disease attending the First Affiliated Hospital of Xinjiang Medical University from July 2021 to May 2022 were selected and genotyped for PPIA gene rs6850 by direct sequencing.The association of this locus with type 2 diabetes mellitus combined with coronary artery disease was evaluated.Logistic regression was used to analyze the association between the rs6850 locus polymorphism and type 2 diabetes mellitus combined with coronary heart disease and non-diabetic coronary heart disease.The association of different genotypes of the rs6850 locus with underlying information was investigated.Results:(1)HDL-C was higher in the stable angina group than in the ACS group and the ACS+T2DM group,and HDL-C was higher in the stable angina+T2DM group than in the ACS group,and the differences were statistically significant(P<0.05).The level of alcohol consumption in the stable angina group and stable angina + T2 DM group was higher than that in the ACS + T2 DM group,and the difference was statistically significant(P<0.05).(2)The difference of GG genotype distribution of PPIA gene rs6850 locus was statistically significant(P<0.05)among non-diabetic coronary artery disease groups(stable angina group and ACS group)in Xinjiang.(3)The distribution of GG,GA,and AA genotypes and alleles at the rs6850 locus of the PPIA gene between the four groups,between the type 2 diabetes combined with coronary artery disease group and the non-diabetic coronary artery disease group,between the type 2 diabetes combined with coronary artery disease group(T2DM combined with stable angina group and T2DM+ACS group),between the stable angina group(stable angina group and stable angina combined with T2 DM group),and between the ACS groups(ACS group and ACS combined with T2 DM group)were not statistically significant(P>0.05).(4)After adjusting for confounders,logistic regression analysis showed that the rs6850 locus did not show any correlation with type 2 diabetes combined coronary artery disease and non-diabetic coronary artery disease under the dominant model,recessive model and overdominant model(P>0.05).(5)BMI was higher in GA genotype carriers compared with AA and GG genotype carriers(P<0.05).Conclusion:The rs6850 single nucleotide polymorphism of the PPIA gene was associated with genetic susceptibility to coronary artery disease in non-diabetic patients in Xinjiang,but not in type 2 diabetes combined with coronary artery disease.
