| Objective: To compare the osteogenic effect of autogenous tooth bone graft material and Bio-Oss.Methods: 18 SPF-SD male rats were randomly divided into 3 groups: A,B and C,with 6 rats in each group.Incisors were extracted on left side of the maxillary arch.Autogenous tooth bone graft material was implanted in teeth extraction socket of rats in group A,Bio-Oss was implanted in teeth extraction socket of rats in group B,any bone graft material was not implanted in teeth extraction socket of rats in group C.The rats were sacrificed 3 months later and were observed with the visual observation,histological observation,serological observation.Results: 1.Visual observation: The boundary of the bone defect area in group A was fuzzy,the texture of the probe was hard,the surface was flat;the boundary of the bone defect area in group B was clear,the fibrous connective tissue can be seen on the surface of the Bio-Oss,and the texture of the probe was slightly hard;the boundary of the bone defect area in group C was clear,an obvious collapse was visible,with fibrous connective tissue growing in,and the texture of the probe was soft.2.Histological observation: HE staining results showed that the autogenous tooth bone graft material was absorbed in the form of insect erosion,and which was wrapped by tightly arranged and regular new bone tissue in group A.In group B,there were more fibrous connective tissue around Bio-Oss,and the new bone was less than that in group A.In group C,fiber connective tissue grew into the basal bone,and a small amount of new bone was formed.The arrangement of new bone tissue was loose and the arrangement of bone cells was disordered.3.Histological measurement and analysis: The rate of new bone formation in group A,B,C were(50.74±5.57)%,(33.76±2.32)%,(11.73±3.94)%,respectively.The rate of new bone formation in group A was better than that in group B,both group A and group B were significantly higher than group C(P<0.001).The percentage of residual bone graft material in group A and group B were(9.63±2.83)%,(27.85±4.37)%,respectively,there had a significant difference(P<0.001).4.Serological observation: The serum levels of ALP in group A,B,C were(3.98 ±0.39)ng/ml,(3.49 ± 0.28)ng/ml,(3.42 ± 0.44)ng/ml,respectively.Compared with the group B and group C,the serum levels of ALP in group A had significant difference(P<0.05).The serum levels of ALP in group B were higher than that in group C,but there was no significant difference between group B and group C(P>0.05).The serum levels of BMP-2 in group A,B,C were(0.98±0.07)ng/ml,(0.85±0.08)ng/ml,(0.83±0.15)ng/ml,respectively.Compared with the group B and group C,the serum levels of BMP-2in group A had significant difference(P<0.05).The serum levels of BMP-2 in group B were higher than that in group C,but there was no significant difference between group B and group C(P > 0.05).Conclusions: Both autogenous tooth bone graft material and Bio-Oss are effective bone grafting materials,which can promote the formation of new bone.The short-term osteogenic effect of autogenous tooth bone graft material is better than that of Bio-Oss,but it still needs to be verified by larger sample size and more observation indicators,and the long-term osteogenic effect needs further observation. |
