Objective: LF3 was used to interfere with the primary cells of C57 mice stimulated by IL-1 β,to verify whether LF3 could reduce the inflammation and matrix degradation of primary chondrocytes stimulated by IL-1 β in C57 mice,and to determine whether LF3 had the effect of reducing joint inflammation and delaying cartilage degradation by intraperitoneal injection of LF3 into the model of knee osteoarthritis(Kneeosteoarthritis,KOA)induced by DMM method.Methods:(1)The effects of LF3 on inflammatory factors,matrix metalloproteinases and type II collagen in chondrocytes were detected by real-time quantitative PCR,immunoblotting and immunofluorescence analysis.(2)The model of knee osteoarthritis in mice was established by DMM method,and LF3 was injected intraperitoneally.The microstructure of knee joint in mice was analyzed by imaging Micro-CT scanning,and the thickness of subchondral bone and bone trabecular density were measured to statistically analyze the therapeutic effect of LF3 on osteoarthritis in mice.(3)The effects of intraperitoneal injection of LF3 on cartilage protection and delay of cartilage degeneration in DMM osteoarthritis mice were studied by saffron fast green staining,hematoxylin-eosin(hematoxylin-eosin,HE)staining,OARSI and synovitis score,and immunofluorescence detection of related target proteins.Results:(1)In the cell experiment in vitro,compared with the model group,LF3 significantly improved the genes and proteins of cartilage-related mechanisms,such as metalloproteinases,type II collagen and inflammatory factors in the experimental group(n=3,P<0.05).(2)In the animal experiment in vivo,the expression of related target proteins in the experimental group was significantly different from that in the KOA model group(n=3,P<0.05).(3)Compared with the KOA model group,the OARSI and synovitis scores of knee joint in the experimental group were significantly different from those in the KOA model group(n=3,P<0.05).(4)Micro-CT results showed that compared with the KOA model group,the subchondral bone loss was reduced in the experimental group after intraperitoneal injection of LF3(n=3,P<0.05).Conclusion:(1)In this study,it was found that after LF3 intervention on chondrocytes stimulated by IL-1β,chondrocyte inflammatory factors,matrix metalloproteinases and type II collagen were significantly improved,and the degradation process of chondrocytes was delayed.(2)In animal experiments,it was found that LF3 had a good effect on the degeneration of knee cartilage,the formation of osteophyte,the recovery of cartilage function and synovitis in KOA mice.
