| Objective:To investigate the value of YAP and Survivin in diagnosis,treatment and prognosis evaluation of tongue squamous cell carcinoma(TSCC),and to provide evidence for potential drug treatment targets of TSCC.Method:The expressions of YAP and Survivin in 99 patients with TSCC and 22 patients with para-carcinoma normal mucosa were detected by immunohistochemistry.Chi-square test was used to analyze the differences in the expression of the two proteins in TSCC and para-carcinoma normal mucosa and their correlation with clinicopathological parameters.Binary logistic regression was used to analyze the risk factors of cancer-related adverse events.Log-rank test and Cox univariate and multivariate regression analysis were used to analyze the risk factors for the prognosis of TSCC.Kaplan-meier method was used to draw the survival curve.The test level was α=0.05,and P<0.05 was considered statistically significant.Results:1.The positive expression rate of YAP in TSCC was 46.46%,which was significantly higher than that in para-carcinoma normal mucosa(0%),and the difference was statistically significant(χ2=16.492,P<0.001).2.The positive expression rate of Survivin in TSCC was 56.57%,which was significantly higher than that in normal mucosa(0%),and the difference was statistically significant(x2=13.277,P<0.001).3.YAP expression level was significantly correlated with PNI,T stage,pN stage and clinical stage of TSCC(all P<0.05),but not with gender,age,smoking,drinking,chewing areca nut,pathological grade and LVI(all P>0.05)..4.Survivin expression level was significantly correlated with PNI,T stage,pN stage,clinical stage,pathological stage,and LVI of TSCC(all P<0.05).There was no correlation with sex,age,smoking,drinking,chewing areca(all P<0.05).5.There was a positive correlation between the expression of YAP and Survivin in TSCC(γs=0.612,P<0.001).6.pN stage and YAP expression level were independent risk factors for postoperative tumor-related adverse events in TSCC patients(P<0.05),and other clinicopathological factors such as age,gender,pathological grade were not related to the occurrence of tumor-related adverse events(P>0.05).7.pN stage and YAP expression level were independent risk factors for prognosis of TSCC patients(P<0.05),while other clinicopathologic factors such as age,gender and pathological grade were not associated with prognosis of TSCC patients(P>0.05).Conclusions:1.The expression level of YAP in TSCC is higher than that in para-carcinoma normal mucosa,which is significantly correlated with PNI,T stage,pN stage and clinical stage of TSCC,and may play a promoting role in the occurrence and development of TSCC.2.The expression level of Survivin in TSCC is higher than that in para-carcinoma normal mucosa,which was significantly correlated with pathological grade,PNI,LVI,T stage,pN stage,and clinical stage of TSCC,and may play a role in the occurrence and development of TSCC.3.YAP and Survivin expression are positively correlated in TSCC,suggesting they may have synergistic effects in TSCC,promoting the occurrence and development of TSCC.4.Positive expression of YAP and pN+are significantly correlated with postoperative adverse tumor events and poor prognosis in TSCC patients.YAP expression level and pN stage may be independent risk factors for the prognosis of TSCC patients. |
