| Objective:In this project,blood samples from coroary heart disease chronic stable exertional angina patients with or without vascular calcification were collected to analyze the differences in inflammatory and calcification indexes,so as to provide clinical basis for the exploration of related mechanisms,construct a vascular calcification animal model at the same time,and use Danshen-Sanqi to intervene to observe the efficacy and mechanism of antagonistic vascular calcification,in order to provide new ideas and evidence for anti-vascular calcification research.Methods:1.Serological characteristics study:A total of 40 patients with vascular calcification and non-calcification of cardiac stasis coronary heart disease(chronic stable exertional angina)were included in this study,including 20 patients in the vascular calcification group and 20 patients in the non-calcification group.The general information and clinical data of the two groups were recorded,and blood samples were collected for coronary heart disease(chronic stable exertional angina)vascular calcification serological index detection,and the scores of medical symptoms,blood lipids,Hcy,,blood calcium,blood phosphorus,calcium-phosphorus product and serological indexes of the two groups were compared.2.Study on the experimental effect and preliminary mechanism of Danshen-Sanqi:15 C57BL/6J mice and 45 ApoE-/-mice were evenly divided into normal control group,model control group,Danshen-Sanqi group,rosuvastatin calcium group,45 ApoE-/-mice were fed with high-fat feed to construct a vascular calcification model,and normal saline,normal saline,Danshen-Sanqi,rosuvastatin calcium suspension were given respectively for intervention,and blood lipids,blood calcium,TNF-α,NF-κB,MGP were determined.Alizarin red staining was used to observe calcification,and Western Blot was used to detect the expression of TNF-α,NF-κB and MGP proteins.Results:1.Serological characteristics study:Compared with the non-calcified group,the calcified group had higher TCM syndrome scores,Hcy,,TG,blood calcium,blood phosphorus,calcium-phosphorus product,TNF-α,NF-κB,and HDL-C and MGP(P were all<0.05).2.Study on the experimental effect and preliminary mechanism of Danshen-Sanqi:Compared with the model control group,Danshen-Sanqi can control the weight gain of calcified model mice,reduce serum LDL-C,HDL-C,TC,TG and blood calcium,down-regulate vascular inflammatory factors TNF-αand NF-κB,and up-regulate the expression of calcification inhibitor MGP.The results of alizarin red staining indicated that Danshen-Sanqi could effectively reduce the occurrence of calcified nodules(all P<0.05).Conclusion:The mechanism of vascular calcification in chronic stable exertional angina is associated with increased expression of inflammatory factors TNF-αand NF-κB and decreased expression of calcification inhibitor MGP.Based on this,it was found that the drug pair of Danshen-Sanqi can exert anti-vascular calcification effect in mice by down-regulating inflammatory factors TNF-α,NF-κB,and up-regulating calcification inhibitor MGP. |
