A Controlled Research On The Protective Effect Of FUYUANXINGNAO And HUOXUETONGFU Treatment On Cerebral Hemorrhage In Rats

Objective: Based on the theory of qi deficiency as the cause of stroke,to compare the prevention and treatment effects of Fuyuan Xingnao method and Huoxue Tongfu method on cerebral edema,inflammation and blood-brain barrier injury after cerebral hemorrhage,and to explore the mechanism of intervention and protection,so as to provide a new theoretical basis for the clinical application of Fuyuan Xingnao method in the treatment of stroke.Methods: 1.A randomized and controlled study was conducted in this study.A total of 96 male SD rats were enrolled by blind method.Three parallel control groups were set up: Fuyuan Xingnao group FYXN,Huoxueftongfu group HXTF,Saline group Saline,and Sham group Sham as blank control.There were26 rats in each group of three parallel control groups,and 18 rats in the single sham operation group.The sham operation group only injected collagenase,and the other three groups were established by collagenase injection method.The Fuyuan Xingnao group was given the concentrated agent of stroke Xingnao Fang(red ginseng,panax notoginseng,Radix rhubarb,ligusticum Chuanxiong)by gavage,and the Huoxuetongfu group was given the concentrated agent of stroke Xingnao Fang(Red ginseng removal)by gavage,once a day after the model was made,and the intervention lasted for three consecutive days.2.Behavioral test methods such as Zea-Longa quintiles were used to evaluate the neurological function of rats.The moisture content of brain tissue was determined by wet and dry weight method.HE staining was used to observe the pathological changes of hematoma and surrounding tissue after intracerebral hemorrhage.Immunohistochemistry was used to detect the expression location and Occludin content of Claudin-5 and Occludin around cerebral hematoma after intracerebral hemorrhage.The expressions of HMGB1 and TLR4 were determined by Westernblot.Results:(1)Compared with sham operation group,neurological deficit score of rats in normal saline group was significantly increased(P<0.05).Compared with normal saline group,Huoxue Tongfu group and Fuyuan Xingnao group were lower than normal saline group(P<0.05).(2)HE staining was used to observe the pathological changes of brain tissue in each group.In the sham operation group,a small number of inflammatory cells were observed to aggregate and infiltrate,and the staining of surrounding nuclei and cytoplasm was uniform.In the saline group,swelling and degeneration of cells were observed,accompanied by a large amount of inflammation Cell infiltration.The degree of brain tissue edema and inflammatory cell aggregation and infiltration in Huoxuetongfu group and Fuyuan Xingnao group were relatively reduced,especially in Fuyuan Xingnao group.(3)Westernblot showed that HMGB1 expression in the normal saline group was significantly higher than that in the sham operation group,with statistical significance(P<0.05).Compared with normal saline group,HMGB1 expression in Fuyuan Xingnao group was down-regulated,and the difference was statistically significant(P<0.05).Compared with normal saline group,TLR4 expression in Huoxue Tongfu group and Fuyuan Xingnao group was down-regulated,and the difference was statistically significant(P<0.05).Immunohistochemical results showed that compared with sham operation group,HMGB1 protein expression was increased in normal saline group around cerebral hematoma.Compared with the normal saline group,the expression of HMGB1 protein in Huoxuetongfu group and Fuyuan Xingnao group decreased.Compared with the sham operation group,the expression of Claudin-5 and Occludin in the tissue around cerebral hematoma in the normal saline group decreased.The expression of Claudin-5 and Occludin in Huoxue Tongfu group and Fuyuan Xingnao group was higher than that in normal saline group.Conclusion:(1)Stroke Xingnao formula(Fuyuan Xingnao method)can protect the neurological function of rats with cerebral hemorrhage and improve the muscle strength of the affected side.(2)Both Fuyuan Xingnao group and Huoxue Tongfu group could improve the lesion degree of hematoma and surrounding tissues,protect neurons and improve the damage of blood-brain barrier,and the improvement was more obvious in Fuyuan Xingnao group.(3)Diarrhea was the main adverse reaction of stroke Xingnao formula(Fuyuan Xingnao method).The adverse reaction was obvious in Fuyuan Xingnao group,and the weight loss of rats was more obvious than that in other groups;No obvious adverse reactions were observed in the Huoxue Tongfu group,and the changes in body weight of rats were not significant compared with the normal saline group.(4)In terms of reducing cerebral edema after cerebral hemorrhage,reducing inflammatory response,and protecting blood-brain barrier,Fuyuan Xingnao method was significantly superior to Huoxue Tongfu method.
