Purpose:Preliminary discussion on patients with thyroid papillary carcinoma patients,thyroid nodules and intestinal flora composition characteristics and differences between healthy people,and explore with and without cervical lymph node metastasis of papillary thyroid carcinoma patients with intestinal flora structure and species abundance.Methods:Fecal samples from 25 patients with thyroid nodules(group A),20 patients with thyroid papillary carcinoma(group B),including 9 patients with cervical lymph node metastasis(Group C)and 11 patients without metastasis(Group D),20 healthy people(Group H)were collected.16 S r RNA was used for gene sequencing of V3-V4 segment of intestinal flora.After sequencing,data results were filtered and spliced and OTU was generated by clustering according to 97% sequence similarity.The species diversity and species difference between groups were analyzed by PCo A,PLS-DA,lefse and other analysis methods,and the flora function was predicted by PICRUSt software.Results:1.There was no significant difference in general information and biochemical examination results between groups;2.Intestinal flora among groups A,B and H α There is difference in β Diversity is different;3.The results of analysis of strain composition show that at the phylum to genus level,the composition of the three groups of dominant flora is similar,and the abundance of some species is different.Compared with group H,the abundance of Actinobacteria,Collinsella,Christensenella,Mogibacterium,Saccharibacteria and Faecalibacterium decreased significantly in group B(P<0.05);The abundance of Parvimonas,Escherichia,Anaerococcus and Holdemania increased significantly(P< 0.05).Compared with group A,the abundance of Akkermansia decreased(P<0.01)in group B,but the abundance of Firmicutes,Verrucomicrobia,Roseburia,Blautia and Holdemania increased significantly(P<0.05)in group B.4.Lefse analysis showed that 8 species,23 species and 31 species were found in group A,group B and group H,respectively;5.Intestinal flora between groups C and D difference in β diversity;6.The composition of dominant flora in group C was similar to that in group D from door to genus level,and different genera were found at the genus level.The abundance of Dorea and Blautia in group C increased significantly(P<0.05),while the abundance of Lachaospira,Parvimonas,Odoribacter and Akkermansia in group D increased significantly(P<0.05);7.Lefse analysis showed that 2 and 9 species of bacteria with significantly increased abundance in groups C and D;8.KEGG function prediction analysis results show that there are differences in 14 metabolic pathways at the second level between group A and group H,11 metabolic pathways at the second level between group B and group H,and multiple metabolic pathways at the third level and level between group A vs group H and group B vs group H.Conculsion:The intestinal flora composition of patients with thyroid papillary carcinoma and cervical lymph node with cancer metastasis is different from that of healthy people.Some of the flora abundance changes,and some of the flora abundance changes may participate in the occurrence and development of thyroid papillary carcinoma.
