| Objective: To determine the expression levels of Mcl-1,Noxa,Bax and Bak in muscle tissues of patients with dermatomyositis,and to investigate their role and significance in the pathogenesis of dermatomyositis.Methods: The expression levels of Mcl-1,Noxa,Bax and Bak in muscle tissues of DM patients and controls were assessed by immunohistochemistry.Results: The expression of Mcl-1 in muscle tissue of the group of patients with dermatomyositis was higher than that of the control group(P<0.05),while the expression of Noxa,Bax and Bak was lower than that of the control group(P<0.05).The expression of Mcl-1 in muscle tissue of dermatomyositis complicated by interstitial lung disease group was higher than that of dermatomyositis without interstitial lung disease group(P<0.05),while the expression of Noxa was lower than that of dermatomyositis without interstitial lung disease group(P < 0.05).The expression of Mcl-1 in muscle tissue of the group of patients with dermatomyositis was positively correlated with the total disease activity,muscle disease activity and lung disease activity of the Myositis Activity Visual Analogue Assessment(MYOACT)(P< 0.05),while the expression levels of Noxa,Bax and Bak were negatively correlated with the total disease activity,muscle disease activity and lung disease activity of the MYOACT score(P < 0.05).Conclusion: Mcl-1,noxa,Bax and Bak proteins may be involved in dermatomyositis pathogenesis through mitochondrial apoptosis,which may be due to the increase of the expression level of anti apoptotic protein Mcl-1,the decrease of the expression level of Pro apoptotic proteins Noxa,Bax and Bak,the decrease of apoptosis of immune cells and inflammatory factors through mitochondrial pathway,and the enhancement of inflammation and immune response,Thus,the muscle tissue lesions of patients with dermatomyositis are aggravated and the disease state persists. |
