Abstract/Summary:
Objective:The expression of SIRT2,P65,Bcl-2,Bax,Caspase-3 and Survivin protein in breast cancer was detected,and the relationship between sirt2,p65,bcl-2,bax,caspase-3 and survivin protein and clinicopathological features of breast cancer was discussed.Methods:The expression of SIRT2,P65,Bcl-2,Bax,Caspase-3 and Survivin in 384 cases of invasive breast cancer and 36 cases of fibroadenosis were detected by immunohistochemical En Vision two-step method.Results:1、The high expression rate of SIRT2 protein in breast cancer(63.8%)was higher than that in fibroadenosis(3.2%)(P<0.05).The high expression rate of Bcl-2 protein in breast cancer(67.6%)was significantly higher than that in fibroadenosis(3.2%)(P<0.05).The high expression rate of Survivin protein in breast cancer(50.8%)was higher than that in fibroadenosis(9.7%)(P<0.05).The high expression rate of Caspase-3 protein in breast cancer(13.0%)was lower than that in fibroadenosis(64.5%)(P<0.05).There was no significant difference between the high expression rates of P65 and Bax protein in breast cancer and fibroadenosis(P> 0.05).The ratio of apoptosis protein Bax/Bcl-2 in breast cancer(0.15)was significantly lower than that in fibroadenosis(3)(P< 0.05).2、The high expression rate of SIRT2 protein in TNM stage Ⅲ-Ⅳ is high,with more lymph node metastasis and higher expression when Ki67≥20%.In molecular typing,the high expression rate of SIRT2 protein in Luminal B1 type is the highest(P<0.05).The expression of other SIRT2 proteins had no significant difference with age,menstrual status and histological grade(P> 0.05).3、The high expression rate of P65 protein in TNM stage Ⅲ-Ⅳ,histological gradeⅠand lymph node metastasis were higher(P<0.05).In molecular typing,the high expression rate of P65 protein in Luminal B1 was the highest(P<0.05).The expression of P65 protein in breast cancer had no statistical significance with age,menstrual status and Ki67 proliferation index(P> 0.05).4、The expression rate of Bcl-2 protein in TNM stage Ⅲ-Ⅳ was high,and the expression rate of grade I in histopathological grade was significantly higher than that in grade II and III(P<0.05),and the expression rate of Luminal A type was the highest in molecular typing(P<0.05).The expression of Bcl-2 protein had no statistical significance with age,lymph node metastasis,menstrual status and Ki67 proliferation index(P> 0.05).5、The expression of Bax protein was low when Ki67≥20%(P<0.05),and the expression of Bax protein was the lowest in HER-2 positive type(P<0.05).The rest had no statistical significance with pathological grade,patient age,lymph node metastasis,TNM stage and menstrual status(P> 0.05).6、The expression of Caspase-3 protein was low when Ki67≥20%(P<0.05),and it was the lowest in Luminal A breast cancer(P<0.05).There was no significant difference with other pathological grades,patient age,lymph node metastasis,TNM stage and menstrual status(P> 0.05).7、The expression of Survivin protein was highest in TNM stage Ⅲ-Ⅳ,in histopathological grade I,when Ki67≥20%,and in molecular typing,the expression of HER-2 was the highest(P<0.05).There was no significant difference between the expression and age,lymph node metastasis,TNM stage and menstrual condition(P>0.05).8、SIRT2 protein was positively correlated with P65 protein,Bcl-2 protein and Survivin protein,but negatively correlated with Bax protein and Caspase-3 protein(P<0.05).P65 protein was positively correlated with Bcl-2 protein and Survivin protein,but negatively correlated with Bax protein and Caspase-3 protein(P<0.05).The expression of Bcl-2 protein was positively correlated with that of Survivin protein,but negatively correlated with that of Bax protein and Caspase-3 protein(P<0.05).The expression of Bax protein and Caspase-3 protein was positively correlated(P<0.05).9、According to the bioinformatics analysis of GEPIA database,the expression of SIRT2,P65 and Bcl-2 in 1085 cases of breast cancer was up-regulated compared with that in normal tissues.According to the analysis of 1271 cases of breast cancer in TCGA database,SIRT2 protein was positively correlated with P65 protein(r=0.13,P<0.05),positively correlated with Bax protein(r=0.28,P< 0.05)and negatively correlated with Caspase-3 protein(r=-0.27,P< 0.05).Conclusions:The up-regulation of SIRT2 protein expression in breast cancer may affect the changes of apoptosis-related proteins in the apoptosis process.May the ratio of apoptosis protein Bax/Bcl-2 decreased,the expression of apoptosis-executing protein Caspase-3 decreased,and the expression of apoptosis-inhibiting protein Survivin increased in breast cancer,thus inhibiting the occurrence of apoptosis in breast cancer cells,and affecting the histological grade and TNM stage of breast cancer,lymph node metastasis,Ki67 proliferation index,molecular typing and other clinicopathological features.This study laid a preliminary experimental foundation for SIRT2 to become a potential target for breast cancer treatment,and also provided a reference for later SIRT2 in vitro experiments in breast solid tumors.
