| Backgroud and Objective: P63 is a member of the p53 tumor-suppressor gene family with structural homology to p53. But they show both similarities and differences in functions. P53 has been widely accepted as an tumor suppressor gene , however, whether p63 is an oncogene or a tumor suppressor gene is still under debate. Inhibitors of apoptosis proteins (IAP) are a family of negative regulators of apoptosis. Survivin is one of IAP family proteins that are expressed during mitosis in a cell-cycle- dependent manner. Survivin may play an important role in regulating chromosome segregation and cell division at the mitotic checkpoint control. Survivin is virtually undetectable in most normal adult tissues, but it is highly expressed during embryogenesis and in most human cancers. The overexpression in malignant tumors has been associated with an aggressive behavior and poor prognosis of the patients. In this study we detected the expression of p63 protein and Survivin protein in non-hodgkin's lymphoma (NHL)and lymph node reactive proliferation with immunohistochemistry to explore the significance of p63 and Survivin in non-hodgkins lymphoma.Materials and Methods: 60 cases of NHL and 10 cases of lymph node reactive proliferation (as control) were collected as study objects. Immunohistochemistry SP staining was adopted to detect the expression rates of p63 protein and Survivin protein in the paraffin wax samples of lymphoma and lymph node reactive proliferation. We did the statistics analysis with x~2 test and rank correlation. Results :1. P63 protein was not expressed in lymph node reactive proliferation.2.The expression rates of p63 protein in NHL was 33.3% (20/60cases) , including DLBCL 71.4% (15/27 cases), MCL 50.0% (1/2 cases), T-LBL 28.6% (2/7 cases), FL 33.3% (2/6 cases).There were negative expressions in MALT PTCL ALCL and MZL. The expression rates of p63 protein in NHL significantly differed from which in lymph node reactive proliferation (p<0.05)3. The positive expression rates of Survivin protein was 40%(4/10 cases) in lymph node reactive proliferation.4.The positive expression rates of Survivin in NHL was 71.1% (43/60 cases) , including DLBCL 81.5% (22 /27 cases), FL 33.3% (2/6 cases), MCL 50.0% (1/2 cases), T-LBL 85.7% (6/7 cases), MALT 42.9% (3/7 cases), PTCL 85.7% (6/7 cases ) ALCL 100% (2 /2 cases) MZL 50.0% (1/2 cases). There was significant difference in the expression rates of Survivin protein between NHL and lymph node reactive proliferation (p <0.05)5. P63 protein and Survivin protein were coexpressed in 13 cases of DLBCL, 2 cases of FL, 2 cases of T-LBL.There was no correlation between the expression rates of p63 protein and Survivin protein in NHL (p>0.05) .6.There were no correlation between the expression of p63 protein and Survivin protein with age sex LDH clinical stage and group extra-nodal extension IPI. But there were significant differences in the expression of p63 protein and Survivin protein between indolent lymphoma groups and aggressive lymphoma groups.7.The clinical therapeutic effects were evaluated in 55 patients whose clinical datas were available after no less than two chemotherapy.The CR rates and PR rates of positive groups of p63 protein significantly differed from the negative groups of p63 protein (p< 0.05). There were significant differences in CR rates between positive groups and negative groups of Survivin protein (p<0.05).Conclusion:1. There were significant differences in the expression of p63 protein and Survivin protein between NHL and lymph node reactive proliferation, so to detect the expression of both proteins has clinical value to discriminate the two diseases.2. There were significant differences in the expression of p63 protein and Survivin protein between indolent lymphoma and aggressive lymphoma , so to detect the expression of p63 and Survivin has clinical value to estimate the malignant degree of lymphoma.3. The expressions of p63 protein and Survivin protein had no correlation to clinical features,but they had close correlation to the clinical therapeutic effects in NHL. The CR rates of positive expression groups was lower than those of negative expression groups respectively, It is significant to estimate the clinical therapeutic effect and prognosis by detecting the expression of p63 and Survivin. |
