Objective:To investigate the expression of miR-27a-3p and PD-L1 in breast cancer tissue and their relationship with clinical pathological features and analyze the correlation between miR-27a-3p and PD-L1.Methods:Collect 40 cases of breast cancer tissue and adjacent breast tissue(≧2cm from the edge of the cancer tissue)and related clinical case data.The expressions of miR-27a-3p in breast cancer tissue and adjacent breast tissue was detected by real-time fluorescence quantitative PCR.The expressions of PD-L1 in breast cancer tissue was detected by real-time fluorescence quantitative PCR and Western blotting.Immunohistochemistry was used to detect PD-L1 protein expression in breast cancer tissue samples.T test was used to analyze the difference in the expression of miR-27a-3p in breast cancer tissue and adjacent breast tissue.The chi-square test was used to analyze the relationship between the expression of miR-27a-3p,PD-L1 in breast cancer and clinicopathological parameters.Spearman correlation analysis was used to analyze the correlation between miR-27a-3p and PDL1,and P<0.05 was considered to be statistically significant.Results:1.The expression of miR-27a-3p in breast cancer tissue and adjacent breast tissue had a significant difference(P <0.001),and the miR-27a-3p in breast cancer exhibited low expression.2.The expression of miR-27a-3p in breast cancer was significantly associated with tumor size,lymph node metastasis,TNM stage,and histological grade(P <0.05).The expression of miR-27a-3p in breast cancer was not related to the age of onset and molecular type(P>0.05).3.The positive expression rate of PD-L1 in breast cancer was 70%.The expression of PD-L1 in breast cancer was significantly associated with tumor size,lymph node metastasis,TNM stage,molecular diced(P <0.05).The positive expression rate of PD-L1 was highest in Luminal B type and lowest in Luminal A type.The expression of PD-L1 in breast cancer was not related to the age,histological grade(P> 0.05).4.The expression of miR-27a-3p mRNA in breast cancer was negatively correlated with the expression of PD-L1 mRNA(r:-0.323,P <0.05).The expression of miR-27a-3p mRNA in breast cancer was negatively correlated with PD-L1 protein levels(r:-0.389,P <0.05).Conclusions:1.miR-27a-3p was low expressed in breast cancer tissue and high expressed in adjacent breast tissue.It was suggested that miR-27a-3p may be involved in the occurrence and development of breast cancer.2.The low expression of miR-27a-3p in breast cancer was significantly correlated with large tumor,lymph node metastasis,high TNM stage,and high histological grade.It was suggested that miR-27a-3p may be related to the growth,proliferation and metastasis of breast cancer and affect the prognosis of breast cancer patients.3.The positive expression of PD-L1 in breast cancer was significantly related to the large tumor,lymph node metastasis and high TNM stage.The expression of PDL1 was significantly different among different molecular types,with the highest positive rate in Luminal B type and the lowest positive rate in Luminal A type.It was suggested that the expression of PD-L1 was related to the prognosis of breast cancer patients,and molecular typing should be considered in the selection of anti-PD-L1 immunotherapy.4.The expression of miR-27a-3p in breast cancer was negatively correlated with the expression of PD-L1,suggesting that the expression of PD-L1 in breast cancer may be related to the expression of miR-27a-3p.
