Abstract/Summary:
Objective:This paper intends to reveal the expression and function of NLRP1 in thyroid cancer through thyroid cancer-related data in the cancer database.It is planned to use immunohistochemical experiments to further study the expression and clinical correlation of NLRP1 inflammasome in PTC.This study can provide new clues for the prevention and treatment of PTC.Methods:1.By analyzing the thyroid cancer cases in the USA Cancer Genome Atlas(TCGA)database,comparing the expression of NLRP1 in thyroid cancer and adjacent normal thyroid tissue,the expression of NLRP1 in the two was analyzed,such as patient age,gender,race,clinical stage,and pathological type was further analyzed.The Kaplan-Meier survival analysis was used to obtain the relationship between the expression of NLRP1 in thyroid cancer and the overall survival of patients.2.The function and pathway of NLRP1 gene: By elucidating the biological function of NLRP1 in thyroid cancer through Gene Ontology(GO)functional enrichment.The Kyoto encyclopedia of genes and genomes(KEGG)was used to analyze the signaling pathway of NLRP1 in thyroid carcinoma.3.By collecting paraffin sections of pathologically diagnosed PTC from January 1,2018 to June 30,2020 in the Department of Pathology,Affiliated Hospital of Guilin Medical College,the tissue more than 2 cm from the tumor edge was called paired paracancerous tissue.The adjacent normal thyroid tissue was used as the control group.The expression level of NLRP1 in thyroid papillary carcinoma was detected by immunohistochemical experiment,the expression difference of NLRP1 between cancer tissue and adjacent normal thyroid tissue was compared,and the relationship between the expression level of NLRP1 in thyroid papillary carcinoma and clinical factors was analyzed.Results: According to the thyroid tissue analysis data in the TCGA database,the expression of NLRP1 in thyroid carcinoma and adjacent normal thyroid tissue was not significantly different(P>0.05).The study showed that the Asian thyroid carcinoma population had higher expression than Caucasians and African Americans,and the difference was statistically significant(P<0.001).The expression level was related to the pathological type of thyroid carcinoma.The expression level of NLRP1 in PTC was higher than that in follicular thyroid cancer(P<0.05).The expression of NLRP1 was not associated with the survival rate of thyroid carcinoma(P>0.05).The function of NLRP1 includes inflammatory response,apoptosis,cell wall acyl dipeptide reaction and innate immune response;KEGG pathway enrichment of NLRP1 suggests that its main pathway is NOD-like receptor pathway.The results of immunohistochemical experiments showed that NLRP1 was highly expressed in papillary thyroid carcinoma tissues,and negatively expressed in adjacent normal thyroid tissues.In addition,experiments showed that NLRP1 expression was associated with tumor size and lymph node metastasis.The expression of NLRP1 was not related to gender,age and clinical stage.Conclusion:The NLRP1 inflammasome is highly expressed in PTC,and its expression level is related to tumor size and lymph node metastasis,suggesting that it may be involved in the occurrence and development of PTC.
