Study On Effection Of Buyang Huanwu Decoction On Epithelial-mesenchymal Transition Induced By Bleomycin In Rats With Pulmonary Fibrosis

Objective:The study explored Buyang Huanwu decoction(Yilin Gaicuo,written by Wang Qing ren)’s influence on epithelial mesenchymal transition in rats which endotracheal injection of bleomycin induced lung fibrosis based on the theory of pulmonary paralysis in Yellow Emperor’s Canon of Medicine,and verified it whether to regulate mi R-21、PTEN/Akt/m TOR signaling pathway.In order to provide theoretical guidance for the pathogenesis of pulmonary fibrosis and provide effective basis for the treatment of TCM syndrome differentiation.Methods:1.Purchase healthy male SD rats(Specific pathogen Free,SPF),all of the rats received5%BLM(5 mg/kg)by endotracheal injection to establish a pulmonary fibrosis model.2.Healthy SD rats were randomly divided into four groups:blank control group,model control group,PFD group,and Buyang Huanwu decoction group.3.Set the mold day to day 0,on the second day of modeling,the saline,PFD suspension,and the Buyang Huanwu decoction were used to treat the correspondinng group of rats,observe the rats’s general status and write down the changes.4.The serum and lung tissues of rats were taken after the 28th、35th、42thday of modeling,weight the lung and calculate the lung quotiety,The serum of HYP,Col I and ColⅢwere tested by ELISA assay,the m RNA expression of E-cadherin、Vimentin、α-SMA、mi R-21 were tested by RT-q PCR,the protein level of PTEN、p-Akt、p-m TOR were tested by Western-blot.Results:1.Rats’s general status after the days of modeling:in the blank control group,rats were active and had good spirit.However,in the other groups,rats’s respiratoryrate became faster than before,their fur became dark and gloomy,and they started having arched backs,they had cyanosis in the outer edges of their limbs and lips,their lung quotiety and the expression of HYP、ColⅠ、ColⅢwere increased(P<0.05 or P<0.01).Additionally,the situation of the rats in the Buyang Huanwu decoction group was better than that in the model control group,their expression of HYP、ColⅠ、ColⅢwere decreased(P<0.05 or P<0.01).2.In the model control group,their m RNA expression level of E-cadherin was lower than blank control group,and their m RNA expression level ofα-SMA and Vimentin was higher than blank control group,Differences were considered significant if P<0.01.In the Buyang Huanwu decoction group,their m RNA expression level of E-cadherin was higher than model control group(P<0.01),and their m RNA expression level ofα-SMA(P>0.05)and Vimentin(P<0.01)was lower than model control group,their m RNA expression level of E-cadherin、Vimentin、α-SMA compared with the PFD group was not significant if P>0.05.3.The m RNA expression of mi R21 in model control group was higher than blank control group,the protein expression level of p-Akt、p-m TOR in model control group was higher than blank control group,the protein expression level of PTEN in model control group was lower than blank control group,Differences were considered significant if P<0.01.The protein expression level of PTEN in Buyang Huanwu decoction group was higher than model control group(P<0.05),the m RNA expression of mi R-21 in Buyang Huanwu decoction group was lower than model group(P<0.01),the protein expression level of p-Akt、p-m TOR in Buyang Huanwu decoction group was lower than model group(P>0.05);with the PFD group,Differences were considered significant if P<0.05.Conclusion:1.Buyang Huanwu decoction could relief the rats’s symptom of pulmonary fibrosis.2.Buyang Huanwu decoction could play anti-pulmonary fibrosis through inhibition epithelial-to-mesenchymal transition.3.The study demonstrated that Buyang Huanwu decoction could inhibit the EMT through mi R-21、PTEN/Akt/m TOR signaling pathway to curb pulmonary fibrosis.
