| Objective:To observe the changes of tissue morphology and the expression of TNF-ɑ and TGF-β in the hippocampal CA1 area of Vascular dementia(VD)rats after acupuncture on the emotional area.To explore the mechanism of acupuncture on the emotional area in the treatment of VD,and to provide experimental evidence for clinical treatment of VD patients.Methods:70 rats were randomly selected for this experiment and divided into 4groups: blank group,sham operation group,model group and treatment group.The water maze test was performed before modeling,and the rats that met the inclusion criteria started this experiment,with a total of 64 rats.The blank group was not given any treatment,the sham operation group only exposed the arterial triangle,and separated the common carotid artery and the vagus nerve.Neurological damage test and water maze test were performed one week after operation,and the experimental data of each group of rats were recorded.In the treatment group,according to Mr.Sun Shentian’s experience in acupuncture,rats were selected for electroacupuncture in the emotional area,and the frequency of 2Hz/5Hz was selected for sparse and dense waves.The current intensity is 1-2m A,the treatment time is 9:00-10:00 am every day,30 min each time,and the cycle is 14 days.After treatment,the Morris water maze test was performed on the rats in each group.After the test,the brains of the rats in each group were collected.The morphological manifestations of the rats in each group were observed by HE staining,and the expression and content of TNF-ɑ and TGF-β were observed by immunohistochemistry and western blot.Results:1.Neurological damage score: Compared with the sham operation group,the difference in neurological damage scores of the rats in each group was extremely significant(p<0.01);compared with the model group,the neurological damage scores of the treatment group were decreased,The difference was significant(p<0.05).2 Behavioural test: Compared with the sham operation group,the learning and memory ability of the rats in the model group was decreased,the escape latency was prolonged(p<0.01),and the number of crossing platforms was decreased(p<0.05);compared with the model group,the rats in the treatment group were The escape latency was shortened(p<0.01),the number of crossing platforms was increased(p<0.05),and the level of learning and memory was improved.3.HE staining results:(1)Model group: neurons in the CA1 region of the hippocampus of rats were damaged,with incomplete structure,unclear edges,irregular shape,disordered arrangement,significant degree of interstitial edema,pyknosis and hyperchromatic nuclei,and hippocampal cells.With significant grants,neuronal loss was evident.(2)Treatment group: In comparison,the neurons in the CA1 region of the hippocampus of the rats were relatively clear and complete in structure,regular in shape,relatively uniform in arrangement,lower in interstitial edema than in the model group,and reduced in nuclear pyknosis.Meta loss is reduced.4.Immunohistochemical results:Compared with the sham-operated group,the expressions of TNF-ɑ and TGF-β in the hippocampal CA1 region of the model group were increased,and the difference was statistically significant compared with the sham-operated group(P<0.01).Compared with the model group,the expressions of TNF-ɑ and TGF-β in the hippocampal CA1 region of the treatment group were both decreased,and the difference was statistically significant between the treatment group and the model group(P<0.01).5.ELISA resultsCompared with the sham operation group,the contents of TNF-ɑ and TGF-β in the hippocampus of the rats in the model group increased(P<0.05);compared with the model group,the contents of TNF-ɑ and TGF-β in the hippocampus of the rats in the treatment group decreased(P<0.05).6.western Blot results:Compared with the sham-operated group,the expressions of TNF-ɑ and TGF-β in the hippocampal CA1 region of the model group were increased,and the difference was statistically significant compared with the sham-operated group(P<0.01).Compared with the model group,the expressions of TNF-ɑ and TGF-β in the hippocampal CA1 region of the treatment group were both decreased,and the difference was statistically significant between the treatment group and the model group(P<0.01).Conclusion:Acupuncture on the emotional area of rats with vascular dementia can reduce the expression of TNF-ɑ and TGF-β proteins.Reducing the occurrence of inflammatory response can protect brain neurons,and ultimately improve the learning and memory ability of vascular dementia rats. |
