Abstract/Summary:
Objective:To investigate the effect of inhibition of inflammatory response by activating Wnt/β-catenin signaling pathway on postoperative cognitive dysfunction(POCD)induced by sevoflurane in elderly rats.Methods:Sixty male rats were randomly divided into three groups(n=20): control group(C group),3% sevoflurane group(S group),lithium chloride(Li Cl)+3% sevoflurane(Li Cl+S group).Group S and Li Cl+S were given 3% sevoflurane inhalation for 6h.Group C was given air for 6h.Li Cl+S group was given Li Cl2mmol/kg intraperitoneal injection once a day for 5 consecutive days before sevoflurane inhalation.Twenty-four hours after sevoflurane anesthesia,10 rats in each group were randomly selected for Morris water maze experiment to test the cognitive function of elderly rats.The navigation experiment was conducted on the first four days,and the space exploration experiment was conducted on the fifth day.The rats were sacrificed immediately after the water maze experiment,and the pathological morphology was observed under a microscope after HE staining.The remaining 10 rats in each group were sacrificed immediately 24 h after sevoflurane anesthesia,and the protein expression levels of TNF-α,IL-1β,β-catenin and p-GSK-3β in hippocampus were detected by Western blot.The expression of β-catenin and p-GSK-3β in hippocampus were detected by immunohistochemistry.Results:Compared with group C,the escape latency of group S and Li Cl+S was significantly longer on the 2nd,3rd and 4th day of water maze experiment(P< 0.05),the residence time in the quadrant where the platform was in the 5th day was significantly shortened(P<0.05),and the number of crossing platform significantly decreased(P<0.05),histopathological damage was found in hippocampal tissue,and the expression levels of TNF-α and IL-1β in hippocampus increased(P<0.05),β-catenin and p-GSK-3βprotein expression levels were decreased in group S(P<0.05),β-catenin and p-GSK-3βprotein expression levels were increased in group Li Cl+S(P <0.05).Compared with group S,the escape latency of the 2nd,3rd and 4th day of the water maze in group Li Cl+S was significantly shortened(P<0.05),the residence time in the quadrant where the platform was in the 5th day was significantly prolonged(P<0.05),the number of crossing platform increased significantly(P<0.05),hippocampal histopathological damage was improved,and the expression levels of TNF-α and IL-1β protein in hippocampus were decreased significantly(P<0.05),β-catenin and p-GSK-3β protein expression levels increased(P<0.05).Conclusion:Li Cl can inhibit inflammatory response and improve postoperative cognitive impairment induced by sevoflurane by activating Wnt/β-catenin pathway.
