Objective:Tra2β(Transformer-2 protein homolog beta)is a serine/arginine-rich-like protein splicing factor and is now implicated to have wide-ranging roles in gene expression as an RNA-binding protein.In this study,we detected the the expression of Tra2β mRNA and Tra2β protein in oral and squamous cell carcinoma(OSCC)cells.Then the Tra2βexpression was knocked down to further clarify its effect of the biological characteristics such as proliferation,migration,invasion and apoptosis of OSCC cells.We aim to explore new targets for the treatment of OSCC.Methods:The qRT-PCR and Western blot were used to detect the expression of Tra2β mRNA and protein in human oral keratinocyte(HOK)and OSCC cells(Cal27,HN6).When Tra2β interfering RNA(si-Tra2β)and negative control(NC)sequence were constructed,we used qRT-PCR to screen the target sequence with the best interference effect and the appropriate concentration,then the Western blot was used to further determine the interference effect.After transfection of si-Tra2β and NC sequence into Cal27 and HN6 cells,the changes of cell proliferation,migration,invasion and apoptosis were detected by colony formation assay,cell wound scratch assay,transwell cell invasion assay and flow cytometry.Results:1.The relative expression of Tra2β mRNA and Tra2β protein was higher in OSCC cells compared with HOK.2.The relative expression of Tra2β mRNA and Tra2β protein was significantly decreased when transfecting si-Tra2β into the OSCC cells.3.Transfection of si-Tra2β into OSCC cells significantly inhibited the proliferation,migration and invasion of OSCC cells,meanwhile,promoted the trend of their apoptosis.Conclusions:1.The expression of Tra2β was increased in OSCC cells.2.Knocking down the expression of Tra2β significantly inhibited the proliferation,migration and invasion of OSCC cells in vitro,meanwhile,promote their apoptosis to a certain extent.
