| Objective:We detected the levels of SOD,MDA and HIF-1α in peripheral blood mononuclear cells(PBMCs)of asthmatic patients,analyzed the correlation between oxidative stress imbalance of PBMCs and HIF-1α level,and explored the role of oxidative stress imbalance of PBMCs and HIF-1α in the pathogenesis of asthma.Methods:From March 2021 to September 2021,a total of 30 patients(13 males and 17 females)who met the inclusion criteria for outpatient treatment and hospitalization of respiratory department of our hospital during acute asthma attack were enrolled as asthma group,and20 healthy subjects(8 males and 12 females)from our hospital during the same period were selected as control group.None of the patients had used hormones for 4 weeks.5ml of elbow venous blood was collected from all subjects and placed in heparin anticoagulant tube.PBMCs were separated by Ficoll density gradient centrifugation method and collected.PBMCs were added into cell cryoprecipitation solution and stored in a refrigerator at-80℃.The levels of SOD,MDA and HIF-1α in PBMCs were detected by ELISA,and lung function indexes(FEV1%pred,FEV1/FVC),p O2,eosinophil count,Ig E value and asthma control test(ACT)score were recorded.Results:1.The levels of HIF-1α and MDA in PBMCs in asthma group were significantly higher than those in control group,with statistical significance(P < 0.05).The SOD level in PBMCs in asthma group was significantly lower than that in control group,the difference was statistically significant(P < 0.05).2.The level of HIF-1α in PBMCs of asthma group was negatively correlated with SOD(P < 0.05),and positively correlated with MDA(P < 0.05).3.The levels of HIF-1α and MDA in PBMCs were negatively correlated with FEV1%pred,FEV1/FVC in asthmatic patients(P < 0.05).4.Oxygen partial pressure was negatively correlated with HIF-1α and MDA levels in asthmatic patients(P < 0.05).5.Eosinophil count in asthmatic patients was significantly higher than that in control group(P < 0.05),and was positively correlated with Ig E value,HIF-1α and MDA level(P< 0.05),and negatively correlated with FEV1%pred,FEV1/FVC,p O2,ACT score,and SOD level(P < 0.05).Conclusion:1.Compared with the healthy control group,the levels of SOD and MDA in PBMCs in asthmatic patients decreased,and there was oxidative stress imbalance in PBMCs in asthmatic patients.The levels of HIF-1α in asthmatic patients were higher than those in healthy subjects,and increased levels of HIF-1α were involved in the course of asthma attack.2.Oxidative stress imbalance of PBMCs in asthmatic patients and HIF-1α jointly promote the development of asthma,and the higher the level of HIF-1α,the more severe the degree of oxidative damage,the lower the antioxidant capacity of asthma patients.3.With the decrease of oxygen partial pressure,the levels of HIF-1α and MDA increased.FEV1%pred and FEV1/FVC decreased with the increase of HIF-1α and MDA,and the degree of airflow obstruction increased.4.Eosinophil count in asthmatic patients was higher than that in healthy control group,and was positively correlated with Ig E value,HIF-1α and MDA level,and negatively correlated with FEV1%pred,FEV1/FVC,p O2,ACT score and SOD level.Oxidative stress imbalance of PBMCs and eosinophils and HIF-1α are involved in the pathogenesis of asthma,aggravating the degree of airflow obstruction in asthma. |
