| Objective:To investigate the effect of butylphthalide on necroptosis mediated by TNF-α/TNFR1 pathway after cerebral ischemia-reperfusion in rats.Methods:Thirty adult male SD rats were randomly divided into 3 groups: Sham group,I/R group and NBP group.The Longa thread method was used to establish cerebral ischemia-reperfusion model in the I/R group and NBP group,while the rats in the Sham group were only separated blood vessels.24 h after reperfusion,the neurological deficit was assessed by the Longa score,cerebral infarction volume was measured by TTC staining,the brain tissue morphology was observed by HE staining,and protein expression levels of TNF-α,RIPK1,RIPK3 and p-MLKL were detected by immunohistochemistry,and the differences among groups were compared.Results:Butylphthalide treatment can reduce the neurological deficit score,the percentage of cerebral infarction volume,the destruction of brain tissue morphology,and the protein expression levels of TNF-α,RIPK1,RIPK3 and p-MLKL in rats after cerebral ischemiareperfusion,and the differences were statistically significant.Conclusion:Butylphthalide therapy can alleviate cerebral ischemia-reperfusion injury,which may be associated with inhibition of necroptosis mediated by TNF-α/TNFR1 pathway. |
