Traditional Chinese medicine(TCM)has a long history and is a fusion of ancient sages’ ideological wisdom,cultural traditions and medical experience.With the national support for the inheritance and innovation,TCM has received wider attention.The research on the material basis of TCM is the key to comprehend scientifically TCM’s efficacy and pharmacological action,and it is also one of the main contents of the modernization research of TCM.In this thesis,the four herbs honeysuckle,licorice,radix isatidis and bitter almond,which are more common and used in large quantities in Chinese herbal medicines,are analyzed by the screening method of database comparison.Comparing the active substances of the two commercially available TCM oral liquids with the above four Chinese medicinal materials as the main raw materials,through statistical analysis,database comparison and other screening methods,the difference information in the two TCM oral liquids has been preliminarily obtained.By supplementing the database and examining the differences in the original formulations of TCM,the research on the differences of the compound extracts was completed,which provided new technical ideas for the optimization of the compatibility and processing of TCM.Firstly,in view of the limitation of the existing database,this thesis established a structure database based on flavonoids with literature research and glycoside binding methods,which covering 2304 compound structures,including 1649 reported structures and 655 derived structures.Based on this database,a retrieval method for chemical constituents in Chinese herbal medicine extracts was established.Secondly,17 different kinds of flavonoid standard mixture solutions were used to optimize the liquid phase separation conditions,including flavonols,flavanes,flavanones,isoflavones and chalcones compounds.The structures and liquid phase retention of the 17 compounds were quite different.And choose the optimal separation conditions to make the separation effect meet the requirements of qualitative analysis.At the same time,the 17 compounds were used to verify the established database.After verification,it was found that the self-built database and the setting of analysis parameters could assist in the accurate identification and judgment of the compound composition in the mixed solution.Using this search method,the composition of the single-party Chinese medicinal materials were analyzed,the initial traceability were completed,and four new flavonoid structures were discovered.In this thesis,based on multivariate statistics,a screening method of significant difference markers was established,and the method was used to analyze two kinds of TCM oral liquids selected in the market.Firstly,through the comparison of pretreatment,the detection method of direct injection was selected,which reduced the process error.Secondly,by comparing the results of statistical analysis,it was found that there were obvious differences in the extraction rate of active substances under different compatibility conditions.At the same time,through the screening of differential markers,a variety of differential substances were found.Finally,combined with the rapid identification method of compounds and the screening method of significant difference markers,in view of the large difference in the extraction rate of four Chinese herbal medicine-related active substances in the two commercially available TCM oral liquids,differences in the composition of the compound extracts before and after the compatibility of the four Chinese medicinal materials with gypsum were compared.Through traceability analysis,it was found that the gypsum components in the formula have a greater impact on the active components of licorice and honeysuckle.The presence of gypsum can greatly improve the extraction rate of chlorogenic acid and other active components from honeysuckle,while inhibiting the active components of licorice such as glycyrrhizin.In order to further study the role of other substances in TCM gypsum in the extraction of medicinal materials,the same analytical method was used,and calcium sulfate was added as a control to extract medicinal materials,and it was finally confirmed that other substances in TCM gypsum had little effect on the extraction effect of active ingredients.The analysis method comprehensively utilized liquid-mass analysis technology,self-built compound database resources and statistical analysis methods,and provided new research ideas and technical support for the research of TCM formulations.
