Traditional Chinese Medicine (TCM) has been used to treat diseases through multi-pathways and multi-targets under the theory of traditional Chinese medicine by the ancient Chinese medicine doctors. The validity of TCM has been proved in the thousands of years'practices. The character of traditional Chinese Medicine is based on an overall analysis of symptoms and signs, the cause, nature and location of the illness and patient's physical condition, which makes TCM attract more increasing attention. However, there are still some problems to solve in the development of TCM, such as, the pharmacological mechanism remains unclear, the effect componets are ambiguous the quality standard is imperfect, which becomes the bottleneck of modernization and development of TCM.In recent years, with the improvement of science and technology, many theories about the effect componets and the quality control of TCM have been proposed by experts at home and aboard. Meanwhile, more precise analytical instruments have been applied in the research of TCM. TCM should be developed deeper using modern theory and analysis method to better ensure the quality of TCM and render it safe, controllable, effective and stable.In present dissertation, according to the characteristics and requirements of quality control of traditional chinese medicine, traditional chromatography techniques have been used to improve the quality standard of Shujinzhitong Tincture (SJZTT)and Tubiechong Medicine(TBC). A mehod based on the LC/MS has been established to find the effective substances of Qifu Decoction(QFD)in vitro and in vivo as well as the xenometabolome of the extract of Fuzi were investigated through LC/MS and Multiple statistical analysis.1. Elevate the quality control standards of Chinese Medicine Prescription and animal drugs(1) Elevate the quality control standard of SJZTTSet up the TLC identification method of Radix Angelicae Pubescentis, add the limit test of Radix aconiti kusnezoffii. The blank contrast showed that the Mateia Medica were non-interferring, this method is easy and simple to handle with a good specificity and repeatability. The method for determination of naringin by HPLC in the prescription was established while there is no such verification item before. (2) Elevate the quality control standard of the TBCSet up the microscopical chatacteristics method of Eupolyphaga sinensis Walker and Steleophaga plancyi(Boleny). Establish the method for determination of the Uracil and Allantoin simultaneously by HPLC in TBC. There were only appearance description and other inspection item in it's former standard. Building this quality control method elevate the quality control level and could be used as reference while revising the quality control standard of other animal drugs.2. Studies on the chemical substances from QFD in vitro and vivoA HPLC-DAD-TOFMS method was applied to separation and identification of compounds in QFD and rat plasma after oral administration. 46 components including 25 diterpenoid alkaloids, 11 flavonoids and 10 triterpenoids were found in dosed rat plasma, and then identified through the matching of empirical molecular formulae with those of published components in an in-house formula library,and further elucidated using dynamic adjustment of the fragmentor voltage to generate structure-relevant fragment ions. After oral administration of QFD, 32 compounds including 28 absorbed components and 3 potential metabolites in rat plasma were detected and identified by comparing and contrasting the compounds measured in QFD with those in the plasma samples by HPLC-TOFMS. The result was an offer of assistance for the quality control and curative mechanism study of QFD3. Study on xenometabolome of the extract of Fuzi in rat urineWe developed a method based on HPLC-TOFMS method coupled with multiple Statistical Analysis technique, which could be applied in screening and identification of TCM xenometabolome in vivo. Based on this method, 34 constituents of xenometabolome from the extract of FZ was screened and identified, including 24 parent components and 10 potential metabolites.
