Study Of Three Drugs In Traumatic Proliferative Vitreoretinal Lesions In Rabbits

Abstract/Summary:
Objective:In this study,we observed the effects of Triamcinolone Acetonide(TA),Artesunate(ART),and Luteolin(LU)in the prevention and treatment of traumatic proliferative vitreoretinopathy after injection into the vitreous cavity of rabbits with traumatic proliferative vitreoretinopathy.Effectiveness.Methods:Forty-eight cyanotic blue rabbits were selected to prepare TPVR animal models by making a penetrating eye injury and intravitreal injection of 0.3 m L platelet-rich plasma,and were randomly divided into four groups(n=12),in which the vitreous cavity of the control group was injected with 0.1 m L saline;the vitreous cavity of the TA group was injected with 0.1 m L(1 mg/m L)triamcinolone acetonide;the vitreous cavity of the ART group was injected with 0.1 m L(20 μg/m L)artesunate;0.1 m L(10 μg/m L)luteolin was injected into the vitreous cavity of the LU group.The vitreous and retinal proliferation were observed by fundus photography and ocular ultrasound at 1,2,3 and 4 wk postoperatively.The expression levels of α-SMA and VIM protein in the vitreous fluid of each group of rabbit eyes were detected by Western Blot at 28 d postoperatively,and the retinal tissue structure of each group was observed by retinal HE staining.Results:1.Results of vitreous and retinal changes in each group: 28 days postoperatively,the mean grades of traumatic PVR in the control,TA,ART and LU groups at 28 days postoperatively were 4,1.08,1.75 and 2.5,respectively.the grade of traumatic PVR in the drug-injected group was significantly lower than that in the control group(P < 0.001),and the difference between the TA group and the ART and LU groups was statistically significant(P < 0.001).The difference between ART and LU groups was not statistically significant(P > 0.05).Fundus images and ultrasound results showed that preretinal proliferative membrane and retinal detachment were seen in the control group;vitreous clouding,preretinal membrane formation and local traction in the LU group;intravitreal proliferative strips and no traction retinal detachment in the ART group;and no proliferative membrane and retinal detachment and only vitreous clouding in the TA group.2.Protein detection in vitreous cavity: At 28 days postoperatively,the differences in the expression levels of α-SMA and VIM protein in the vitreous of the four groups were statistically significant(P < 0.001).α-SMA and VIM protein expression levels in the vitreous fluid of the TA,LU and ART groups were lower than those of the control group,and α-SMA and VIM protein expression levels in the vitreous fluid of the LU and ART groups were higher than those of the TA group,and α-SMA and VIM protein expression levels in the vitreous fluid of the ART group were higher than those of the LU group.-SMA and VIM protein expression levels in the vitreous fluid of the ART group were higher than those of the LU group,and the differences were statistically significant(P < 0.01).3.HE staining of retinal tissues: At 28 days after surgery,the arrangement of retinal layers in the control group was disordered,severely distorted or locally fractured,the structure of each layer was unclear,the anterior membrane was obviously thickened,and the retinal nerve layer was obviously detached;LU group:the arrangement of retinal layers was slightly distorted,inflammatory exudation was visible in front of the retina,and superficial retinal detachment;ART group: the structure of retina was clear,mild edema,and superficial detachment was visible;TA group: the structure of retinal layers was clear,the arrangement was still neat,local retinal folds were visible,and there was no retinal detachment..Conclusion:Intravitreal injection of tretinoin,artesunate and lignocaine were all effective in preventing and treating traumatic PVR,with tretinoin being the most effective.
