Backgrounds Endotheline (ET) is a kind of anti-injury factor which plays the important role in the tissue reparation of human body. Proliferative vitreoretinaopathy (PVR) after rhegmatogenous retinal detachment is a series of cellular events that include dedifferentiation, migration, proliferation, and production of extracellular matrix (ECM), followed by membrane formation and traction along surfaces of the detached retina resulting in failure of surgical retinal reattachment. Retinal pigment epithelial cells (PRE) are major proliferative cells in PVR, Furthermore, the imbalance of growth factors, such as TGF,TNF in eye, also play an important role in PVR. ET enchances the motosis of cells, but its effects on the periretinal membrane (PRM) has been unknown. Besides, it facilitates the contraction of cells by touching the receptors to stimulate the contrction of a-Smooth muscle actin (a-SMA). a-SMA is one of the components of cytoskeleton, located in cell plasma of different types of cell, It is essential for the normal growth, maturation, differentiation, intergrity and function of its cells, and related to migration and traction of cells which contribute to the contraction of PRM. But the relationship between ET and a -SMA in the PRM of PVR has not been reported. The infrasound is a kind of 0.0001 -20 Hz unaudible sonic waves surrounding our environment which can induce various biologic effects on many parts of human body such as CNS, auditory system, heart,lung and liver to some extents. Nowadays, the research on the infrasound is rare; the research on ERG, EOG, YEP to SD rats shows that not only does it indirectly introduce the abnormality of vision through CNS, but it influences the conduct of NS by the abnormality of blood fluid to the eye as well. ET is a kind of nervous repairing factor, and its expression on the injury of retina up-regulates. However, the expression of ET to the infrasonic injury of retina has not been published.Purposes CD To exame the expression of the ET-1 in PRM in order to discuss its effect on P\Tl. (2) To observe the expression of the ET-1 and a -SMA in PRM in order to discuss the effect of two in PVR. (3) To discuss the expression of ET-1 in retina of Sprague-Dawley rats after exposure to infrasound.Methods (D 14 pieces of the PRMs obtained from 14 cases of PVR of retinal deachment (RD) undergoing vhretomy were used to exame the expression of ET-1 and a - SMAby immuno-histochemical detection. ㎝ale Sprague-Dawley rats were exposed to infrasound of 16 Hz~130 dB for 2 hours everyday. The rats were killed after exposure of 1 day, 7 days, 14 days and 21 days respectively and immunochemistry detection was used to study the expression of ET-1 in retina of SD rats.Results ?The positive staining showed in some cells and extracellular matrix (ECM) in the different grades of the PRMs. And the stains was deepened at the rim of PRM in which some of cytonucleus was become the shape of fusiform and/or slimer and longer. (2) The reaction of ET-1 showed in the cells and ECM in the various grades of the PRMs and darken at the rim of the PRM in which some of cytonucleus became the shape of fusiform and/or slimer and longer. While a -SMA expressed in the centra and verge of various grades of the PRM, and the positive cells parall to the contractive orientation of the PRM. ?The expression of ET-1 of retina changed after exposure of 7 days and 14 days. But ET-1 was dominantly expressed in outer retinal segment(ORS) after 7 days, expressed in inter retinal segment(TRS) after 14 days. By the time of exposure to infrasound, the injury in retina gradually extended from ORS to IRS.Conclusions ?It hints that the endotheline involves in PVR, and exists in cells and ECM,especially strengthens at the rim of the PRM, which may be related to the constraction of surface of the PRM. (2) The ET-1 and a - SMA all express in the PRM, the cells in the PRMs maybe express ET-1 and concret it to ECM and effect on the contractive cells that express a -SMA.
