| Objective: To explore the role and potential clinical value of differential expression of long non-coding RNA(lnc RNA)and colorectal cancer associated transcript 1(CCAT1)in early screening and diagnosis of epithelial ovarian cancer.Methods: From June 2020 to December 2021,serum samples of patients with epithelial ovarian cancer(EOC)(including EOC,before EOC postoperative chemotherapy and after EOC postoperative chemotherapy)in obstetrics and Gynecology of the Third Affiliated Hospital of Zunyi Medical University(Zunyi first people’s Hospital),patients with benign epithelial ovarian cancer,patients with ovarian cyst and women with normal physical examination in the physical examination center at the same time were collected,Combined with the clinical medical record system and test report system,the clinical data and CEA and CA125 test results of patients were collected.The relative expression level of CCAT1 in serum was detected by real-time quantitative reverse transcription polymerase chain reaction(RT-q PCR).Fisher’s test was used to analyze the relationship between the expression of serum CCAT1 and the clinicopathological characteristics of patients with epithelial ovarian cancer(including age,tumor size,FIGO stage,lymph node metastasis,distant metastasis,etc.).Receiver operating characteristic(ROC)was used to evaluate the diagnostic efficacy of serum CCAT1,CEA and CA125 in epithelial ovarian cancer.Pearson correlation coefficient(PCCS)was used to analyze the correlation between the relative expression of serum CCAT1 and CEA and CA125.Finally,the diagnostic effect of serum CCAT1 combined with CEA and CA125 on epithelial ovarian cancer was evaluated.Results:(1)The relative expression of serum CCAT1 in EOC patients increased,which could significantly distinguish normal control(P<0.001),but it could not distinguish patients with EOC before postoperative chemotherapy,EOC after chemotherapy,epithelial benign ovarian cancer and ovarian cysts(P>0.05);The expression of CCAT1 in patients with stage Ⅲ+Ⅳ EOC [4.177(2.460-6.742)] was higher than that in patients with stage Ⅰ+Ⅱ EOC(P=0.008);The expression of EOC before postoperative chemotherapy(4.771±2.748)was higher than that after postoperative chemotherapy [2.701(1.646-4.258)](corrected P=0.0149);There was no significant difference in the expression of serum CCAT1 between epithelial benign ovarian cancer(3.232±1.604)and ovarian cysts(2.532±1.613)(P=0.2186),but both were higher than normal control(1.138±0.531)(P<0.001).(2)The up regulation of serum CCAT1 expression in EOC patients was positively correlated with FIGO stage(P=0.024),lymph node metastasis(P=0.008),distant metastasis(P=0.008)and pelvic and peritoneal effusion(P=0.024),but not with age(P=1.000)and tumor size(P=0.359).(3)The AUC of serum CEA,CA125 and CCAT1 in normal control group and stage I+II EOC group were 0.6821,0.8750 and 0.8152 respectively;The AUC of normal control group and stage Ⅲ+Ⅳ EOC group were 0.5978,1.0000 and 0.9658respectively;The AUC of stage I+II EOC group and stage Ⅲ+Ⅳ EOC group were0.5402,0.6786 and 0.8393 respectively;In terms of diagnostic efficacy,CA125 is better than CEA and CCAT1 in distinguishing between normal control and stage I+II EOC,normal control and stage III+IV EOC,but not as good as CCAT1 in distinguishing stage I+II EOC and stage III+IV EOC,suggesting that CCAT1 is better than CEA and inferior to CA125 in early EOC early warning,but it can better indicate the prognosis of patients.(4)Pearson correlation coefficient analysis showed that the relative expression of serum CCAT1 in EOC patients had no correlation with CEA and CA125(r~2=-0.182,P=0.417;r~2=0.055,P=0.809).(5)The sensitivity and specificity of CCAT1 combined with CA125 in detecting EOC were 86.36% and 100.00% respectively.The comprehensive diagnostic efficiency was higher than that of CCAT1 alone or combined with CEA.Conclusion: The expression of lncRNA CCAT1 in the serum of EOC patients is significantly increased,which has a good early diagnostic efficacy for EOC.It is expected to become an independent tumor marker for early diagnosis and judgment of the disease condition of EOC.The combined detection of lnc RNA CCAT1 and CA125 can improve the comprehensive diagnostic efficacy for EOC. |
