Objective: To detect the expressions of long non-coding RNA of colon cancer associated transcript 1(lncRNA CCAT1)and long non-coding RNA(lncRNA)PTV1 in the normal endometrial tissues and the endometrial carcinoma tissues,to analyze the relationship between the expression level and the clinicopathological parameters of endometrial carcinoma,and to explore their role in the occurrence and development of endometrial carcinoma.Methods: Forty cases of normal endometrial tissues and forty cases of endometrial carcinoma tissues were collected.Real time-PCR was employed to test the expression of lncRNA CCAT1,lncRNA PVT1 in different endometrial tissues,the relationship between their expression and clinicopathological parameters of endometrial cancer was analyzed.To investigate the role of lncRNA CCAT1 and lncRNA PVT1 in the occurrence and development of endometrial carcinoma and its significance in the early diagnosis,clinical treatment and prognosis of endometrial carcinoma.Results:1 The relative expression of lncRNA CCAT1 in endometrial carcinoma was 1.2096(0.2042,7.6889),the relative expression in normal endometrial tissues was 0.2342(0.0846,0.07947),the expression in endometrial carcinoma tissues was significantly higher than that in normal endometrial tissues(P<0.05),there were significant differences between the two groups.The relative expression of lncRNA CCAT1 in endometrial carcinoma tissues of stage ?+? was higher than stage ?+?,in patients that myometrial invasion?1/2 was higher than that<1/2,in patients with lymph node metastasis was higher than that without lymph node metastasis(P<0.05).The expression of lncRNA CCAT1 in different age,pathological grades,hypertension,diabetes,hyperlipidemia,obesity,whether ER-positive,whether PR-positive of endometrial carcinoma patients had no statistical difference(P>0.05).2 The relative expression of lncRNA PVT1 in endometrial carcinoma was 1.8349(0.1617,15.0387),the relative expression in normal endometrial tissues was 0.2805(0.0507,1.0729),the expression in endometrial carcinoma was significantly higher than that in normal endometrial tissues(P<0.05),there were significant differences between the two groups.The relative expression of lncRNA PVT1 in endometrial carcinoma tissues of stage ?+? was higher than stage ?+?,in patients that myometrial invasion?1/2 was higher than that<1/2,in patients with lymph node metastasis was higher than that without lymph node metastasis,and the difference was statistically significant(P<0.05).The expression of lncRNA PVT1 in different pathological grades of endometrial carcinoma had no statistical difference(P>0.05).The expression of lncRNA PVT1 in different age,hypertension,diabetes,hyperlipidemia,obesity,whether ER-positive,whether PR-positive of endometrial carcinoma patients had no statistical difference(P>0.05).Conclusions:1 lncRNA CCAT1 was highly expressed in endometrial carcinoma,and its expression increased with the depth of myometrial invasion,lymph node metastasis and stage.2 The expression of lncRNA PVT1 was up-regulated in endometrial carcinoma,and its expression level was closely related with stage,myometrial invasion depth and lymph node metastasis.3 lncRNA CCAT1 and lncRNA PVT1 may promote the occurrence,development,invasion and metastasis of endometrial carcinoma by regulating the target gene.They are can be used as a new tumor marker for the diagnosis and treatment of endometrial carcinoma.
