Objective: Through the method of network pharmacology,screen the main effective components of Jiawei Wuhu Zhuifeng powder in the treatment of Parkinson’s disease,speculate the target of its effective components,construct the traditional Chinese medicine component target disease interaction network,and explore the action mechanism of Jiawei Wuhu Zhuifeng powder in the treatment of PD,so as to provide a scientific and theoretical basis for in-depth experimental research.Methods: TCMSP and TCMID were used to retrieve and collect the chemical composition information of six traditional Chinese medicines in Jiawei wuhuzhuifeng powder,including cicada molting,tiannanxing,Gastrodia elata,scorpion,Bombyx Batryticatus and earth palm root.The names of target proteins were standardized with the help of uni prot database,Construct the potential target data set of the effective components of Jiawei wuhuzhuifeng powder.Through gene cards database,dis Ge net database and CTD database,the targets of Parkinson’s disease related diseases were collected and screened,the target information data set of the active components of Jiawei Wuhu Zhuifeng powder was constructed,and the common targets of drugs and diseases were obtained by Venn diagram.Using the visual network software Cytoscape 3 8.0 construct the component target disease network of modified wuhuzhuifeng powder in the treatment of Parkinson’s disease.Obtain the high confidence interaction relationship of common target proteins in string database,in Cytoscape 3The protein interaction network(PPI)was constructed in 8.0 software,and the cyto NCA plug-in was used to analyze the topology of the network,so as to screen the core target of modified Wuhu Zhuifeng powder in the treatment of Parkinson’s disease.Go enrichment analysis and KEGG pathway enrichment analysis were carried out for potential targets and signal pathways through metascape database.Results:(1)289 active components of Jiawei Wuhu Zhuifeng powder were collected through tcmsp database and tcmid database,including 16 active components of cicada molting,123 active components of tiannanxing,46 active components of Gastrodia elata,16 active components of scorpion,10 active components of Bombyx Batryticatus and 78 active components of earth palm root;There are 1340 potential targets of modified wuhuzhuifeng powder.(2)8514 targets related to Parkinson’s disease were obtained through gene cards database,dis Ge net database and CTD database.(3)According to Venn diagram,there are 202 targets of active components of traditional Chinese medicine of modified Wuhu Zhuifeng powder and PD disease intersection targets,of which the main targets are AKT1,actb,INS,Jun,CASP3,etc.(4)2618 go entries were enriched by go enrichment,including 2305 biological process entries,121 cell function entries and 192 molecular function entries.The molecular functions mainly include DNA binding transcription,RNA polymerase II specific factor binding,DNA binding transcription,ubiquitin like protein and so on,which mainly occur in membrane raft,membrane micro domain,synaptic membrane and so on.(5)Through the enrichment analysis of KEGG pathway,a total of 190 pathways were obtained,including lipid and atherosclerosis signaling pathway,age-rage signaling pathway,IL-17 signaling pathway,thymoid hormone signaling pathway,etc.Conclusion(s): The stigmasterol contained in the modified wuhuzhuifeng powder such as β Sitosterol,quercetin,oleic acid,methyl eugenol and other core components,act on key factors such as AKT1,ACTB,INS,Jun and CASP3,then regulate age-rage signal pathway,IL-17 signal pathway,thymoid hormone signal pathway,lipid and atherosclerosis signal pathway,inhibit heterologous stimulation response,regulate cellular nutrition level,metal ion level and cellular oxidative stress response,Finally,it plays a role in the treatment of PD.
