Objective:Using data mining to explore the clinical experience of Professor Tan Zihu in the treatment of Parkinson’s disease,summarize the law of prescription and medication,and summarize his medication experience.The core prescription obtained from data mining is analyzed through network pharmacology to speculate its mechanism of action in the treatment of Parkinson’s disease,so as to provide a basis for the clinical verification of the efficacy of traditional Chinese medicine in the next step.Method:1.Collect the data of Parkinson’s disease patients in the outpatient department of Hubei traditional Chinese medicine hospital treated by Professor Tan Zihu from January 2020 to October2021,use Microsoft Excel 2016 to enter the basic information and prescriptions of patients,establish a database,standardize the data,use the ancient and modern medical case cloud platform software for data processing,and count the frequency,four Qi,five flavors,meridian tropism and efficacy of traditional Chinese medicine,Excavate the commonly used drugs,drug pairs and core prescriptions to explore the prescription and medication law of Professor Tan Zihu in the treatment of Parkinson’s disease.2.Collect and retrieve the effective chemical components and corresponding target information of rose,Uncaria and Cistanche deserticola through tcmsp,retrieve the target information not found by tcmsp through batman-tcm database,and retrieve the effective components and corresponding targets of Dendrobium,Pugongying and Gastrodia elata through batman-tcm database.Through the drugbank database and genecards database,the known disease targets related to Parkinson’s disease were searched with the keywords "Parkinson’s disease" and "Parkinsonism".The intersection of the two genes and the target of the disease is input into the PPI database to obtain the intersection of the two genes.The biological process analysis of target gene ontology(GO)and signal pathway enrichment analysis of Kyoto Encyclopedia of genes and genomes(KEGG)were carried out on the top 50 core target genes through David database to illustrate the role of target proteins in gene function,and Cytoscape 3 9.0 software to construct compound target pathway network.Result:1.the results of this study showed that the incidence rate of female was slightly higher than that of men,and the age distribution was mostly concentrated at 50-79 years old.According to the results of drug use frequency,the drugs with the highest frequency include Dendrobium,rose,dandelion,Cistanche deserticola,uncaria,Salvia miltiorrhiza,Gastrodia elata,licorice,white peony and keel.According to the statistical results of four Qi,warm and peaceful drugs are used the most,followed by slightly cold,cold,cool and slightly warm drugs.According to the statistical results of five flavors,sweet,bitter and pungent drugs are mainly used,followed by salty,sour,slightly bitter,astringent and light.According to the statistical results of meridians,the drugs of liver meridians are the most,followed by spleen meridians,heart meridians and stomach meridians.From the statistical results of efficacy,it can be seen that the main drugs are heat clearing and detoxification,moistening intestines and defecating,and calming liver yang.The results of association rule analysis show that the drug pairs include Dendrobium rose,Dendrobium dandelion,dandelion rose,Dendrobium Cistanche deserticola and Dendrobium Uncaria.The core prescription is Dendrobium,rose,dandelion,uncaria,Cistanche deserticola and Gastrodia elata.2.The main components of core prescription drugs for the treatment of Parkinson’s disease are quercetin β-Sitosterol,lycopene,m-hydroxybenzoic acid,kaempferol,taraxacinol,3-hydroxycarotene,4-hydroxybenzylamine,taraxacinol,choline,etc;The core targets for the treatment of Parkinson’s disease are:AKT1,INS,IL6,TNF,TP53,IL1 B,CASP3,VEGFA,Jun,CTNNB1;The core prescription treatment of Parkinson’s disease mainly affects the biological processes such as negative regulation of apoptosis and positive regulation of RNA polymerase II promoter transcription by affecting enzyme binding,the same protein binding,transcription factor binding and other ways,acting on cell components such as extracellular space,extracellular area and platelet granule cavity,and targeting tumor necrosis factor,HIF-1,toll like receptor Nod like receptor,MAPK,FOXO,PI3 K Akt,apoptosis and other signal pathways play a role in the treatment of Parkinson’s disease.Conclusion:1.Professor Tan Zihu’s commonly used drugs for Parkinson’s disease include Dendrobium rose,Dendrobium dandelion,dandelion rose,Dendrobium Cistanche deserticola and Dendrobium Uncaria.2.Professor Tan Zihu’s core prescription for the treatment of Parkinson’s disease is: Dendrobium,rose,dandelion,uncaria,Cistanche deserticola and Gastrodia elata.3.The core prescription may act on extracellular space and other cellular components through tumor necrosis factor,HIF-1,toll like receptor,nod like receptor,MAPK,FOXO,PI3 K Akt,apoptosis and other signal pathways,and affect biological processes such as negative regulation of apoptosis,so as to play a role in the treatment of Parkinson’s disease.
