Background:Histone deacetylase 6(HDAC6)is a unique member of class IIb HDAC family.It mainly exists in the cytoplasm.It can participate in the occurrence and progression of liver cancer by regulating various biological processes such as cell proliferation,angiogenesis,apoptosis,autophagy and tumor immunity.It has been proved that hepatitis B virus X protein(HBx)and cytochrome P450 2E1(CYP2E1)are closely related to the occurrence of hepatocellular carcinoma(HCC).At present,there are few studies on the correlation between the expression of HDAC6,HBx and CYP2E1 and the differentiation of HCC.Objective: To explore the correlation between HDAC6 and HBx and CYP2E1 expression in hepatitis B virus related HCC and its clinical significance,so as to provide reference for further elucidate the pathogenesis of HCC.Methods: Collecting the 42 male hepatitis B virus related HCC samples that from the pathology department of the Affiliated Hospital of Yan bian University form 2012 to 2021.H-E staining was used to determine the differentiation degree of all HCC,and divided into the well differentiation group(n = 22)and poorly differentiation group(n = 20).Immunohistochemical staining method was used to observe the expression levels of HDAC6,HBx and CYP2E1 in well differentiated and poorly differentiated HCC cells.And the staining results of HDAC6,HBx and CYP2E1 were scored by hcore scoring method.SPSS software was used to analyze the data,and chi square test was used to analyze the relationship between the different degree of differentiation of HCC and the expression of HDAC6,HBx and CYP2E1;Spearman correlation method was used to analyze the correlation between the expression of HDAC6 and the expression of HBx and CYP2E1 in HCC.Results:1.H-E staining result: 42 cases of hepatitis B virus related HCC specimens were well differentiated HCC accounted for 52.38%(22 cases),poorly differentiated HCC accounted for47.62%(20 cases).2.The results of hcore scoring method: The negative expression of HDAC6 in well differentiated HCC accounted for 9.09%(2 cases),weak positive expression accounted for 50%(11 cases),and positive expression accounted for 40.91%(9 cases);In poorly differentiated HCC,the negative expression of HDAC6 accounted for 25%(5 cases),the weak positive expression accounted for65%(13 cases),and the positive expression accounted for 10%(2 cases).The weak positive expression of HBx in well differentiated HCC accounted for about 27.27%(6 cases),positive expression accounted for 50%(11 cases),and strong positive expression accounted for about22.73%(5 cases);The positive expression of HBx in poorly differentiated HCC accounted for 35%(7 cases),and the strong positive expression accounted for 65%(13 cases).The negative expression of CYP2E1 in well differentiated HCC accounted for about 9.10%(2 cases),weak positive expression accounted for about 45.45%(10 cases),and positive expression accounted for about45.45%(10 cases);The negative expression of CYP2E1 in poorly differentiated HCC was 20%(4cases),the weak positive expression was 65%(13 cases),and the positive expression was 15%(3cases).3.Chi square test results: The high expression rate of HDAC6 in well differentiated HCC tissues was 40.91%(9/22),which was 10%(2/20)in poorly differentiated HCC.The high expression rate of HDAC6 in well differentiated HCC was significantly increased(P<0.05).The expression of HDAC6 was related to the degree of differentiation of HCC.The high expression rate of HBx was72.73%(16/22)in well differentiated HCC tissues and 100%(20/20)in poorly differentiated HCC tissues,and HBx expression was higher in poorly differentiated HCC(P<0.05).The expression of HBx was correlated with the degree of differentiation of HCC.The high expression rates of CYP2E1 in well differentiated and poorly differentiated HCC tissues were 45.45%(10/22)and 15%(3/20)respectively.The expression of CYP2E1 in well differentiated HCC increased significantly(P<0.05).The expression of CYP2E1 was correlated with the degree of differentiation of HCC.4.Spearman rank analysis showed that there was a negative correlation between the expression of HDAC6 and HBx in all hepatitis B virus related HCC(r=-0.483,P<0.05;r=-0.550,P<0.05;r=-0.600,P<0.01);There was a positive correlation between the expression of HDAC6 and CYP2E1 in all hepatitis B virus related HCC(r=0.740,P<0.01;r=0.579,P<0.05;r=0.699,P<0.01).Conclusion:1.The expression of HDAC6 and CYP2E1 in well differentiated HCC was significantly higher than that in poorly differentiated HCC,and the expression of HBx in poorly differentiated HCC is higher than that in well differentiated HCC,suggesting that the expression of HDAC6,CYP2E1 and HBx is correlated with the degree of differentiation of HCC.2.The expression of HDAC6 was negatively correlated with the expression of HBx and positively correlated with the expression of CYP2E1 in HCC,suggesting that the three may play different roles in the occurrence and development of HCC.
