| Xiehuo Xiaoying Recipe is a clinically-experienced prescription that has been used for many years in the Jiangsu Provincial Hospital of Integrated Traditional Chinese and Western Medicine.It has the effects of clearing liver fire,nourishing yin and calming the heart.Early clinical studies have found that Xiehuo Xiaoying Recipe can help patients with Graves’diseases(GD)to quickly improve the function reaching the standard,improving the disease recovery rate,and reducing the application of anti-thyroid drugs(ATDs),has unique advantages in the treatment of GD.In this paper,based on the preliminary clinical efficacy,the mechanism of Xiehuo Xiaoying Recipe in the treatment of GD model mice from the liver is explored;then,HPLC-QTOF/MS was used to study the chemical components of Xiehuoxiaoying formula and the blood components of GD mice.Combining network pharmacology and molecular docking technology,clarify the material basis of its efficacy in preventing GD;further develop it into modern granules,this article carried out the pharmaceutical research of granules(technology research,quality standard research and stability test).The main research contents are as follows:1.Use GD model mice to study the mechanism of Xiehuo Xiaoying Recipe from the liver.After the mice were grouped,modeled,and administered,the secretion levels of T4 and TRAb in the serum of each group of mice were detected,and the pathological changes of thyroid tissue were observed by the HE staining method;then the levels of ALT and AST in the serum of each group of mice were detected and analyzed the expression levels of GSH,MPO and MDA in liver tissues.The results showed that the extract of Xiehuo Xiaoying Recipe significantly inhibited T4 and TRAb in the serum of mice,and inhibited the expression of MPO and MDA in the liver,and increased the expression level of GSH in the liver tissue,and the effect was dose-dependent.It indicates that the extract of Xiehuo Xiaoying Recipe may have a protective effect on liver damage in GD model mice by reducing oxidative stress.2.HPLC-QTOF/MS was used to analyze and identify the chemical components of Xiehuo Xiaoying Recipe extract and the blood components of the extract of Xiehuo Xiaoying Recipe after gavage of GD mice.71 chemical components were identified from extracts of Xiehuo Xiaoying Recipe,and 31 prototype compounds were identified from serum samples.It clarified the main chemical components of Xiehuo Xiaoying Recipe and the prototype chemical components that entered the serum,revealed the composition of the chemical components in vivo and in vitro,and laid the foundation for the follow-up basic research of medicinal substances.3.Based on network pharmacology and molecular docking technology to study the pharmacodynamic material basis and mechanism of Xiehuo Xiaoying Recipe for GD.The chemical composition and target set of Chinese medicinal materials of Xiehuo Xiaoying Recipe were collected through the preliminary mass spectrometry analysis and database,and then the target set related to GD disease was searched.Construct the "medicinal material-chemical composition-target" network and the "target protein-target protein" interaction PPI network of Xiehuo Xiaoying Recipe for the treatment of GD.Carry out KEGG and GO enrichment,predict the relevant mechanism of action,and conduct correlation analysis.Finally,the screened active ingredients are molecularly docked with key targets.Obtained 92 compounds of Xiehuo Xiaoying Recipe and 227 corresponding targets.The results of molecular docking showed that the core components of Xiehuo Xiaoying Recipe have good binding activity with potential key targets,which is similar to the affinity of current drugs that may treat GD.This study revealed the relevant targets and metabolic pathways of Xiehuo Xiaoying Recipe,and initially revealed the effective material basis of Xiehuo Xiaoying Recipe in the treatment of GD.4.The preparation process of Xiehuo Xiaoying Granules was studied.The original prescription was used in the clinic in the form of decoction,so this experiment is based on the form of clinical medication,and the process research is carried out with water extraction.Orthogonal test was used to determine the best water extraction process based on the gentiopicrin content and solid yield index;refer to the gentiopicrin content transfer rate to determine the temperature of the concentrated liquid;combine the process and drug properties to determine the optimal water extraction process;The molding rate is used as an index to screen process parameters such as auxiliary materials,auxiliary material dosage,ethanol concentration,etc.,to determine the best molding process.5.The quality standard and stability of Xiehuo Xiaoying Granules were investigated.In order to establish the quality standard of Xiehuo Xiaoying Granules,the characteristics,identification,inspection and content determination were investigated,and the thin-layer identification method for the three medicines such as gentian,anemarrhena and pansanglong was established as well as the gentian in Xiehuo Xiaoying granules.The method for determining the content of picroside has been formulated as a draft quality standard.Furthermore,the stability of the preparation was investigated,and the initial stability of Xiehuo Xiaoying Granules was investigated by accelerated tests and long-term test methods,and the results showed that the quality of the preparation was stable. |
