| Objective The purpose is to investigate the expression of serum LncRNA HOTAIR,LncBRM,and ICR in the serum of healthy people,patients with liver cirrhosis,and patients with different stages of liver cancer,and compare the sensitivity and specificity of LncRNA HOTAIR,lncBRM,ICR and AFP in the serum expression of liver cancer patients,and further improve the early diagnosis value of serum lncRNAs in primary liver cancer.With the help of modern scientific and technological methods,the expression levels of serum LncRNA HOTAIR,LncBRM and ICR in patients with liver cancer were detected and compared with the TCM syndrome of patients,so as to find out the correlation between the two and provide evidence for clinical syndrome differentiation and treatment.Methods According to the inclusion and exclusion criteria,86 cases of primary liver cancer,20 patients with liver cirrhosis,and 20 healthy patients were selected from our hospital from February 2019 to January 2020.Referring to the 9th edition of "Traditional Chinese Medicine" and the 3rd edition of "Traditional Chinese Medicine Clinical Diagnosis and Treatment of Oncology Specialties",patients with primary liver cancer are classified according to TCM syndrome types:Qi stagnation and blood stasis type,liver depression and spleen deficiency type,liver and kidney yin deficiency type,dampness-heat intrinsic type and dampness-blood stasis mutual type;and according to-the-2017 version of liver cancer diagnosis and treatment,staging is divided into stage Ⅰa,Ⅰb,Ⅱa,Ⅱb,Ⅲa,Ⅲb,and Ⅳ.Collect the discarded serum of all volunteers,determine the serum AFP value by chemiluminescence enzyme immunoassay(CLEIA)method,and determine the expression of serum LncRNA,HOTAIR,lncBRM,ICR by quantitative reverse transcription polymerase chain reaction(qRT-PCR).Results 1.The serum levels of LncRNA HOTAIR,LncBRM,and ICR in PLC patients were significantly higher than those in liver cirrhosis group and healthy group(P≤0.001),and the expressions of serum LncRNAs in liver cirrhosis group were higher than healthy group(P<0.05).2.The serum expression levels of serum LncRNA HOTAIR and LncBRM in the PLC group with late clinical stage were significantly higher than those with early stage(P=0.004,P=0.006),patients with extrahepatic metastasis were significantly higher than those without extrahepatic metastasis(P=0.044,P=0.029),patients with tumor thrombogenesis were significantly higher than those without tumor thrombogenesis(P=0,P=0.001),and the tumor diameter>5cm group was higher than the diameter≤5cm group(P=0.038,P=0.034).3.ROC curve was drawn for PLC diagnosis based on serum LncRNAs level.The area under the LncRNA HOTAIR curve was 0.916,When the expression level of LncRNA HOTAIR was 0.000082,the sensitivity was 86.9%and the specificity was 79.5%.The area under the curve of LncRNA BRM was 0.888,When the expression level of LncRNA BRM was 0.000039,the sensitivity was 82%and the specificity was 79.5%.The area under the curve of LncRNA ICR was 0.92,When the expression level of LncRNA ICR was 0.000294,its sensitivity was 90.2%and specificity was 82.1%.The area under the curve of AFP was 0.804,its sensitivity was 78.7%,and its specificity was 82.1%,suggesting that the expression levels of serum LncRNA HOTAIR,LncBRM and ICR were all higher than AFP in the diagnosis of PLC patients.4.There was no significant correlation between TCM syndromes of PLC patients and serum LncRNA HOTAIR,LncBRM and ICR expression levels.Conclusion Serum LncRNA HOTAIR,LncBRM,and ICR were significantly highly expressed in the serum of PLC patients,moderately expressed in patients with cirrhosis,and not expressed or poorly expressed in healthy patients.The expression of LncRNA HOTAIR and LncBRM in the serum of PLC patients is related to clinical stage,extrahepatic metastasis,vascular invasion,tumor embolism and tumor size,which may be involved in the occurrence and development of PLC.There is no obvious correlation between LncRNA HOTAIR and TCM syndrome types,and there is insufficient evidence for clinical TCM syndrome differentiation.Serum LncRNA HOTAIR,LncBRM and ICR levels have potential diagnostic value for PLC. |
