Background and Objective:Heart failure(HF)is the final stage of various heart diseases.It not only brings a load-bearing burden to society and families,but also has a high mortality and disability rate,which seriously affects the quality of life and prognosis of patients.Therefore,early diagnosis and identification of heart failure is of great significance to the prognosis of patients with heart failure.It is found that non coding micro RNA(miRNA)and long non-coding RNA(LncRNA)are widely distributed in the cardiovascular system,and play an important role in cardiac development,pathological process of heart failure,prognosis and gene regulation network.Studies have found that LncRNA HOTAIR can inhibit myocardial hypertrophy and is a protective factor of the heart.Bioinformatics was used to predict the miRNAs that may bind to LncRNA HOTAIR,and verified by functional analysis.Finally,miR-126-5p was screened.This study detected the expression levels of LncRNA HOTAIR and miR-126-5p in serum of patients with heart failure,analyzed their expression levels and clinical significance in heart failure,and discussed the possible mechanism.Materials and Methods : Selected patients were admitted to the Department of Cardiology of the First Affiliated Hospital of Dali University from July 2021 to November 2021.In addition,59 patients with heart failure in line with this experiment were selected as the control group,including 30 healthy physical examination subjects during the same period.All have signed family informed consent forms.The clinical data were complete,including age,sex,underlying diseases,smoking and other basic data of the control group and the heart failure group.In terms of cardiac function classification,the patients were divided into three groups according to the standards formulated by The New York Heart Association(NYHA): 16,31 and 12 patients with cardiac function in class II,III and IV,respectively.The expression levels of LncRNA HOTAIR and miR-126-5p in the experimental group and the control group were detected by real-time quantitative PCR to explore the clinical significance of serum LncRNA HOTAIR and miR-126-5p in heart failure.Results:1.There were no statistically significant differences in gender,smoking,age,alcohol consumption,ethnicity and diabetes between the heart failure group and the control group(P> 0.05).There were statistically significant differences in BNP value,LVEF,LVEDD and NLR between the heart failure group and the control group(P <0.05).Age,drinking,sex,smoking,coronary heart disease,ethnicity,dilated cardiomyopathy,atrial fibrillation,hypertensive heart disease,P> 0.05,the difference was not statistically significant.2.The relative expression levels of LncRNA HOTAIR and miR-126-5p in the heart failure group were lower than those in the control group,with statistical significance(P< 0.05).3.The relative expression levels of LncRNA HOTAIR and miR-126-5p were positively correlated(P< 0.05).4.The relative expression levels of LncRNA HOTAIR and miR-126-5p were positively correlated with LVEF(P<0.05),and the difference was statistically significant.5.The relative expression of miR-126-5p was negatively correlated with different cardiac function grades and LVEDD(P<0.05),and the difference was statistically significant.6.In the diagnosis of heart failure,the AUC value of miR-126-5p was 0.801,P < 0.05,and the difference was statistically significant.Conclusion: 1.The relative expression levels of LncRNA HOTAIR and miR-126-5p in the heart failure group were lower than those in the control group.2.The relative expression levels of LncRNA HOTAIR and miR-126-5p were positively correlated with LVEF,and the relative expression levels of miR-126-5p were negatively correlated with different cardiac function grades and LVEDD.3.The expression of miR-126-5p can be used as an independent predictor in the diagnosis of heart failure.
