objective: To explore the growth advantages and possible regulatory mechanisms of periplaneta Americana extract on refractory wounds of rats.Methods: the rats were divided into blank group,model group,blattella America group and baifuji group according to the random number table method.The neonatal wound indexes of each group were weighed and compared.The expressions of Smad6 and Smad9 were detected by immunohistochemistry.Histopathological morphology and histopathological score were observed by HE staining.The ultrastructure and mitochondria were observed by transmission electron microscopy.Results: the neonatal wound index of the experimental group was significantly higher than that of the model group and the blank group,and the difference was statistically significant.Immunohistochemistry showed that the expression of Smad6 in the experimental group decreased,while the expression of Smad9 increased,which was statistically significant compared with that of the model group,the blank group and the control group(p<0.05),The inflammatory reaction in pathological morphology group was rapid.Tem showed that the cell structure of the experimental group was closer than that of the blank group,the experimental group and the control group,and the number of mitochondria was increased,which was statistically significant compared with the model group(p<0.05).Conclusion: in the TGF-1/Smads pathway,blattella America extract may accelerate the pathologic inflammatory process of wound cells by down-regulating Smad6 and up-regulating Smad9 expression,promoting the proliferation and differentiation of mitochondria,endoplasmic reticulum and desmosomes,and promoting wound healing. |
