| Objective:To identify the main factors influencing the synthesis of caffeic acid derivatives by analyzing the synthetic methods of caffeic acid derivatives and caffeic acid amide derivatives.To summarize the hypoglycemic effect of different derivatives.Methods:Cinnamic acid phenethyl ester was synthesized by using chemical synthesis method respectively,two kinds of ester derivatives of caffeic acid and caffeic acid amide derivatives of caffeic acid phenethyl ester,summarize condensation reagent,reaction degree yield of the molar ratio of reactants and reaction time on the composition,and for caffeic acid,caffeic acid phenethyl ester and caffeic acid amide derivatives of 45 mice for hypoglycemic effect were studied by analysis of hypoglycemic effect of different drugs.Results:When the reaction temperature was 160 ℃,the yield of cinnamic acid at different reaction time was significantly higher than that of other reaction temperature in the same reaction time(P < 0.05).Under the reaction temperature of 150 and 160 ℃ ℃,the yield of cinnamic acid at the reaction time of 50 minutes can reach the highest value,which is 29.88% and 77.42%,respectively.The yield of cinnamic acid at the reaction temperature of 170 was the highest(31.03%)when the reaction time was 30 minutes,℃and the yield of cinnamic acid was higher than that of other different reaction times at the same reaction temperature(P < 0.05),and the yield of cinnamic acid was 31.03%when the reaction time was 30 minutes.But there was no significant difference compared with other reaction time(P > 0.0).5.Under the conditions of 30 and 40 ℃ ℃,the yield of cinnamic acid reached the highest value,71.33% and 61.14%,respectively,when the amount of thioyl chloride was 2.4 mmol.The yield of thioyl chloride in the same reaction time was significantly higher than that in the same reaction time(P <0.05),but the difference in the other groups was significant(P > 0.05).The optimum reaction conditions of cinnamic acid were condensed and refluxed at 160 for 40 min,℃and the optimum synthetic conditions for phenylethyl cinnamate were 100 ℃oil bath for 120 min.Production of caffeic acid at 140 ℃for 30 min The highest yield was59.93%,followed by 52.31%(40 min),which was higher than that at the same reaction temperature(P < 0.05).The optimum reaction conditions of caffeic acid and ethyl caffeic acid were as follows: condensation and reflux at 140 for 30 min and synthesis ℃of phenethyl caffeic acid in oil bath at 100 ℃for 30 min.The condensation effect of NMM/IBCF was relatively high,and the yield was 63.37,which was significantly higher than that of other condensation reagents under the same reaction material(P <0.05).But there was no significant difference in the yield of different raw materials under the same condensation reagent(P > 0.05).0.05;The increase of the molar amount of caffeic acid can effectively increase the yield of the product,and when the molar amount of caffeic acid is 1 mmol,the molar ratio of amino acid is increased.Arginine and lysine increased significantly,while isoleucine and serine increased first and then decreased.There was no significant difference in blood glucose levels between the two groups at 0 min after 30 minutes,the blood glucose levels of A1A1A2A3A3A3A6A7 and A8 were significantly decreased.Compared with the control group,there was a significant difference(P < 0.05),while A4 and A5 were significantly different from the control group(P < 0.05).The level of blood glucose in the group was basically unchanged,and there was no significant difference(P > 0.05).Conclusion:The yield of caffeic acid derivatives and caffeic acid amide derivatives is affected by different reaction time,reaction temperature and molar ratio of substance.With the exception of caffeic acid serine amide and caffeic proline amide,there was no significant effect on blood glucose,caffeic acid arginine amide.Caffeic acid,phenylalanine amide and other derivatives of caffeic acid amides can play a good role in reducing blood sugar,which should be paid more attention to and popularized. |
