| Objective: To establish a method for determining the the contents of 11 mineral elements in the decoction of three testacean traditional Chinese medicines(Oysters,Abalone shell,Concha arcae)and calcined one,and compare the difference on "calming the liver and suppressing Yang" in rats with hypertension of hyperactivity of liver Yang,and the relationship between mineral elements and nature and efficacy of testacean TCM were preliminarily discussed.To provide the experimental basis for the scientific research and clinical application and new ideas for the development and application of testacean TCM.Methods:1.The samples were digested by microwave digestion system,and the amounts of 11 mineral elements(Ca,Mg,Fe,K,Cu,Zn,Mn,Co,Mo,Se,Cr)were determined by inductively coupled plasma optical emission spectrometry(ICP-OES),and the differences of their contents were compared.2.Spontaneously hypertensive rats(SHR)were fed with Fuzi decoction for 5weeks to replicate the hyperglycemic rat model of liver-yang hyperactivity.Each group were separately treated with oyster decoction,calcined oyster decoction,abalone shell decoction,calcined abalone shell decoction,concha arcae decoction,calcined concha arcae decoction by oral administration.After 2 weeks of treatment,the degree of irritability and conjunctival hyperemia in each group were observed.The 24 h intake and 24 h intake were measured.The systolic pressure(SP)was measured with non-invasive tail artery.Plasma catechol(CA),angiotensin II(Ang II)and levels were measured by ELISA.Left ventricular mass index(LVMI)was measured in rats and hematoxylin eosin(HE)staining was used to observe the morphological changes of rat heart.Results:1.Part one experimental results: The order of the content of each element in the decoction of three testacean TCM has the same trend.The order of the content of inorganic elements in the water decoction of oysters: Ca>Mg>K>Mn> Fe>Cu>Zn>Se(Cr)>Mo(Co);the order of the content of inorganic elements in the water decoction of abalone shell:Ca>Mg>K>Mn>Fe>Zn>Cu>Se(Cr)>Mo(Co);the order of the content of inorganic elements in the water decoction of concha arcae: Ca>Mg>K>Mn>Fe>Cu>Zn>Se(Cr)>Mo(Co).The contents of Mg,Mn,and Fe in the decoction of raw oysters and abalone shell were more than two times higher than those in concha arcae.In the decoction of oysters,abalone shell,and concha arcae,the content of Ca was higher than that of the decoction of each raw product(P<0.05),and there was no significant difference in the contents of K,Se,and Cr in the decoction of each raw product(P>0.05).The contents of Mg,Mn,Fe,Cu,Zn,Mo and Co have a great difference compared with the raw water decoction.2.Part two experimental results:(1)Experiment 1 Results: Compared with WKY group of rats,the model group rats shaggy and lacklustre,easy to anger,bite when fetching,irritability degree level of Ⅲ mostly,eyes combined with membrane congestion degree heavier.In the model group,the amount of water intake increased,the intake decreased,the face temperature increased,the systolic blood pressure increased,and the comparison with the WKY group was statistically significant(P<0.05 or P<0.01).Compared with model group,captopril group rats hair loose lacklustre,but its degree of irritability and eye combined with membrane ease congestion degree(P<0.05).The rats in the oyster group,abalone shell group,and concha arcae group had compact and shiny hair.The degree of irritability was grade I and the degree of conjunctival hyperemia was reduced(P<0.05).Rats in captopril group,oyster group,abalone shell group and concha arcae group decreased water intake,increased food intake,and decreased facial temperature and systolic blood pressure were significantly different from those in the model group(P<0.05 or P<0.01).There were no statistical differences in hair bulkiness,color,irritability,eye conjunctival hyperemia,drinking water intake,food intake,facial temperature,systolic blood pressure between the model group and the calcined oyster group,the calcined abalone shell group and the calcined concha arcae group(P>0.05).(2)Experiment 2 Results: Compared with WKY group,the content of CA,Ang II and ALD in plasma of model group increased significantly(P<0.01),and the serum NO content decreased significantly,and the difference was statistically significant(P<0.01).Compared with the model group,the plasma CA and Ang II levels in the captopril group,oyster group and abalone shell group were significantly reduced(P<0.01),and serum NO levelswere significantly increased(P<0.01).The contents of NE and ALD in plasma in rats of concha arcae group were decreased(P<0.05),and serum NO level were increased(P<0.05),and there was no significant difference in plasma E,Ang II level between the model group and tconcha arcae group(P>0.05).The levels of plasma CA,Ang II,ALD and serum NO in calcined oyster group,calcined abalone shell group and calcined concha arcae group were not significantly different from those in model group(P>0.05).(3)Experiment 3 Results: Compared with WKY group,the rats in the model group did not receive treatment,and the weight loss and LVMI increased significantly(P<0.01).Compared with the model group,the LVMI of captopril group,oyster group and abalone shell group decreased significantly(P<0.01 or P<0.05).The myocardial cells of the rats in the captopril group,the oyster group,the abalone shell group and the concha arcae group were improved.There was no significant difference between the LVMI and the model group in the calcined oysters,the calcined abalone shell group and the calcined concha arcae group(P>0.05).Conclusion:1.The order of the content of each element in the decoction of three testacean TCM has the same trend: Ca>Mg>K>Mn>Fe>Cu or Zn>Se(Cr)>Mo(Co);the content of each inorganic element in decoction of three kinds of Traditional Chinese medicine forged products was different from that of the decoction of each raw product,and only the content of Ca elements increased in comparison with the water decoctions of their raw materials.2.The effect of oysters and abalone shell raw products on liver and Yang is better than that of concha arcae raw materials,which can reduce the blood pressure and improve the general symptoms of rats with hyperactivity of liver Yang,and has a certain protective effect on the heart of Spontaneously hypertensive rats.The mechanism may be related to the regulation of plasma CA,Ang II,ALD levels and serum NO levels.3.The oysters,abalone shell and concha arcae calcined products have no obvious improvement of blood pressure and general symptoms in rats with hyperactivity of liver Yang,and there is no obvious improvement of myocarditis in rats with hyperactivity of liver Yang,suggesting that the three calcined products may have no obvious effect on calming the liver and suppressing Yang.4.The difference in potency of three kinds of raw testacean TCM of oysters,abalone shell and concha arcae may be influenced by the type and content of inorganic elements.The change of the quality of three kinds of testacean TCM may be related to the change of inorganic elements content before and after calcining. |
