This study was performed to investigate the protective role of Schisandra chinensis bee pollen extract(SCBPE)against renal injury and liver injury induced by Cisplatin(CP)in rats and its functionary mechanism and aimed to provide a powerful experimental basis for the comprehensive development and utilization of bee pollen.The main contents and results of this research are summarized as follows:1.Protective effect of SCBPE on liver and kidney injury induced by CP in rats:The rats were fed adaptively for 7 days and divided into six groups of six animals each and treated as the following:control group,model group,positive control group,SCBPE groups(400,800,1200 mg/kg/day,respectively)for 12 consecutive days,and intraperitoneally injected with CP(8 mg/kg)at seventh day,except for control group intraperitoneally injected with saline.Compared with model group,high dosage of SCBPE significantly reduced the kidney index,the levels of BUN,CR,ALT and AST in serum,the contents of MDA and iNOS in liver and kidney,and significantly increased the activities of SOD,CAT,and the content of GSH in liver and kidney.In the model group,the HE staining of liver tissue showed that the hepatic lobules were necrotic,and the hepatic cells were balloon degeneration,accompanied with a large number of inflammatory cell infiltration.The HE staining of renal tissue showed renal tubular lesions in renal tissue,including renal tubular epithelial cell vacuolar degeneration,epithelial cell shedding,inflammatory reaction in the stroma.In the SCBPE administration groups,the pathological changes of liver and kidney in rats were better than those in the model group,which further confirmed the protective effect of SCBPE.2.SCBPE attenuated CP induced inflammation in liver and kidney:Compared with model group,high dosage of SCBPE significantly reduced the levels of TNF-α,IL-6 and IL-18 in serum,the expression of inflammation cytokines(NFκB,IL-6,IL-1β,CD 14 and CD68)in liver and kidney,and then gradually decreased in a dose-dependent after administration of SCBPE.These results indicated that SCBPE was likely though regulating TNF-α and NFκB pathway to inhibit inflammation,thereby exert the protective effect.3.SCBPE attenuated CP induced apoptosis in liver and kidney:Compared with model group,high dosage of SCBPE significantly reduced the expression of apoptosis-associated genes(Bax,P53,caspase3,caspase9,cytochrome C)in liver and kidney induced by CP,and then gradually decreased in a dose-dependent after administration of SCBPE.These results indicated that SCBPE could regulate the apoptosis pathway to inhibit apoptotic,thereby exert the protective effect.In conclusion,SCBPE could reduce the damage of liver and kidney caused by CP by reducing the level of oxidative stress,inflammation and apoptosis,and improving the antioxidant,anti-inflammatory and anti-apoptotic capacity of the body.This study could provide a powerful experimental basis for the comprehensive development and utilization of bee pollen.
