Clinical Study Of Serum SFlt-1 And 25-OH-VD Expression In Pregnant Women With Recurrent Miscarriage And Their Correlation With TCM Constitution

Objective:To study the levels of soluble fins-like tyrosine kinase receptor(Flt-1)and 25 hydroxy vitamin D(25 OH-VD)in peripheral blood of patients with recurrent abortion during early pregnancy.To investigate the expression and significance of serum s Flt-1 and 25 hydroxy vitamin D in early pregnancy of patients with recurrent abortion.Study on the distribution of TCM constitution in 40 cases of recurrent abortion pregnancy.To explore the correlation between different TCM constitutions and serum sflt-1 and 25 hydroxy vitamin D.Follow up the NT outcome after treatment of recurrent miscarriage pregnancy and provide theoretical basis for clinical treatment.Methods:40 cases of recurrent abortion early pregnancy were selected as experimental group and 31 cases of normal early pregnancy(without history of spontaneous abortion)as control group..(1)The enzyme linked immunosorbent assay(ELISA)was used to determine the level of sflt-1 in serum of two groups.(2)Chemiluminescence Microparticle Immuno Assay(CMIA)was used to determine the level of 25 hydroxy vitamin D in the serum of two groups.(3)The expression levels of sflt-1 and 25 hydroxy vitamin D in serum of the two groups were compared and analyzed.(4)Analyze of TCM constitution distribution of 40 patients with recurrent abortion in this study.(5)To investigate the correlation between TCM constitution and serum sflt-1 and 25 hydroxy vitamin D in patients with recurrent abortion.(6)Statistical comparison of NT outcomes in patients with recurrent miscarriage and normal pregnancy.Results:(1)The serum level of sflt-1 in the experimental group(258.56±270.89pg/ml)was significantly higher than that in the control group(107.24±108.30pg/ml),P < 0.05.;(2)The level of 25 hydroxy vitamin D in the experimental group(13.54±4.29ng/ml)was significantlylower than the control group(16.97±6.52ng/ml),P < 0.05;(3)The higher the concentration of25 hydroxy vitamin D,the lower the serum sflt-1 expression level;(4)In the TCM constitution of recurrent abortion patients,Yang deficiency is the main factor(34.3% of total frequency),Yin deficiency(34.3% of total frequency)and Qi stagnation(14.3% of total frequency);(5)In Yang deficiency,the level of serum s Flt-1 expression was(184.85 ±201.63pg/ml),25 hydroxy vitamin D expression level(13.69 ±3.55 ng/ml).In Yin deficiency,the level of serum s Flt-1expression was(179.50 ±138.12pg/ml),25 hydroxy vitamin D expression level(14.11 ±2.20ng/ml).In Qi stagnation,the level of serum s Flt-1 expression was(141.30 ±151.82pg/ml),25 hydroxy vitamin D expression level(14.14 ± 2.62ng/ml).There was no significant difference between groups,P > 0.05;(6)There were 29 cases of normal NT,11 cases of abortion,24 cases of normal NT in normal early pregnancy and 7 cases of abortion.There was no significant difference between the two groups,P > 0.05.Conclusions:(1)The expression of serum s Flt-1 increased in the early pregnancy of the patients with recurrent abortion.(2)The expression of 25 hydroxy vitamin D decreased during early pregnancy in recurrent abortion patients.(3)There is a negative correlation between25-hydroxyvitamin D and serum s Flt-1.(4)The TCM constitution of recurrent abortion patients is mainly yang deficiency.(5)The expression of serum s Flt-1 in patients with Yang deficiency in recurrent abortion is higher than that in other constitutions,the expression level of 25 hydroxy vitamin D is lower than that in other constitutions.(6)Low 25 hydroxy vitamin D and elevated serum s Flt-1 expression may be the causes of repeated pregnancy failure in patients with recurrent abortion.(7)After treatment,it can improve the success rate of re-pregnancy in patients with recurrent abortion,and provide theoretical basis for clinical treatment.
