Objectives:IGT is the early stage of diabetes,is a key stage for reversible normal.This study intends to eliminate hate IGT rat model of Danxiao Tea,to observe the changes of the expression of leptin receptor in kidney and liver adiponectin receptor R2mRNA,to explore the mechanism of treatment of Danxiao Tea to IGT rats,in order to provide new experimental evidence for the prevention and treatment of IGT.Methods:1 The two method(high fat and high glucose diet and intraperitoneal injection of STZ 20mg/kg)IGT rat model was established.The animal were divided into normal group,model group,Chinese medicine treatment group,positive control group and exercise group.Chinese medicine treatment group with dose of Danxiao Tea,the positive control group for two doses a double guanidine,exercise group with swimming training intervention.2.Intervention after 8 weeks,blood was measured by chromatometry,blood glucose measurement automatic-biochemical analyzer,TC,TG,HDL-C,LDL-C,ELI SA method for detection of insulin,adiponectin,leptin.3.In kidney tissue pathological examination;the kidney by immunohistochemical staining of leptin receptor(ob-R)expression;expression of liver by using Realtime RT-PCR method to detect adiponectin receptor 2 mRNA.Result:1.The blood glucose,glycosylated hemoglobin,insulin and so on in the model group were higher than those in the normal group,and the difference was statistically significant2.Lipid changes:compared with the normal group,model group TC,LDL-C were significantly increased(P<0.01),TG(P<0.05)also increased,HDL-C decreased significantly(P<0.01);compared with the model group,TC LDL-C Danxiao Tea group,decreased significantly(P<0.01),TG also decreased(P<0.05),HDL-C increased(P<0.01),metformin group and exercise group TC decreased significantly(P<0.01),TG,LDL-C decreased(P<0.05),the level of HDL-C increased(P<0.05)。3.Fat factor ADPN,the change of LEP level:in IGT model group rats ADPN were significantly lower than the normal group(P<0.01 group),the level of LEP was significantly higher than the normal group(P<0.01);compared with the model group,ADPN significantly increased Danxiao Tea(P<0.01),the level of LEP was significantly decreased(P<0.01).4.Renal pathological model group glomerular matrix hyperplasia,focal inflammatory cell infiltration,glomerular cysts increased obviously,dilatation of tubules,glomerular volume increased slightly;traditional Chinese medicine treatment group the pathological changes of renal tissues in model group is relatively improved.5.Immunohistochemistry results of ob-R:the expression of ob-R in rats of model group was significantly lower than that in normal group(P<0.01);compared with the model group,the positive expression of Danxiao Tea group could obviously increase the ob-R(P<0.01).6.Fluorescence quantitative RNA results adiponectin receptor R2 in hepatic tissue:expression of adiponectin receptor R2 mRNA in model group was significantly lower than that of normal group(P<0.01);compared with the model group,the expression of Danxiao Tea treatment group can significantly increased adiponectin receptor R2 in IGT rats(P<0.01).Conclusions:1.IGT rat model of LEP was significantly higher than that in normal group,while the expression of ob-R in the kidney was significantly lower than normal group,suggesting that IGT stage leptin resistance and ob-R expression of adiponectin R2mRNA abnormalities and IGT lesions related to.2.IGT rat model and hepatic ADPN expression than normal group significantly decreased,and the abnormal expression of IGT and adiponectin R2 on lipid metabolism.3.Danxiao Tea interposing,significantly increased serum levels of ADPN,LEP and blood lipid significantly decreased,the expression of ob-R and adiponectin R2 ignificantly increased,blood glucose HbA1c and FINS were significantly decreased,suggesting Danxiao Tea to have certain influence to improve the degree of pathological changes of IGT.4.Danxiao Tea consumed in IGT rats the treatment mechanism may be by reducing the secretion of LEP,increased ADPN secretion,reduce blood fat,and upregulation of ob-R and adiponectin receptor R2 expression,increased insulin sensitivity the improvement of insulin resistance,thereby reducing the blood glucose,blood lipid.
