| Objective:Under the instruction of traditional Chinese medical theory and clinical experience,this study combine Changshan-huyou with Hawthorn and Cassia-seed respectively.Through comparing the lipid-dropping effect and antioxidant capability of different combination,this study determine the optimal combination and build a solid foundation for drug development.Methods:(1)Through evaluating Soxhlet extraction,ultrasonic extraction,reflux extraction and water extraction four different methods by total flavonoids of hawthorn and extraction yield,we find the optimum extraction process and refine it by orthogonal experiments.(2)Comparing Soxhlet extraction,ultrasonic extraction,reflux extraction and water extraction four different methods by total anthraquinone of cassia seed,the optimum extraction process is determined and then refined through orthogonal experiments.(3)Through feeding high fat diet,hyperlipidemia rat model is established.After six weeks of preventative medication,Serum biochemical indices,body weight,liver weight and liver coefficient of pathological changes are measured.(4)Liver antioxidant ability of hyperlipidemia rats is evaluated through antioxidant index to investigate the effect of different combination of Changshan-huyou drop lipid formula.Results:(1)The optimum extraction process of total flavonoids in hawthorn was ethanol refluxing.The extraction conditions were ethanol concentration 60%,liquid solid ratio 14:1,extraction time 1 h,extraction times 3 times.(2)The optimum extraction process of total anthraquinone in fried cassia seed was ethanol refluxing method.The extraction conditions were ethanol concentration 70%,liquid solid ratio 25:1,80℃ water bath,reflux extraction 2 h.(3)Different formula has different lipid drop effect.① Liver index results show that Huyou group,hawthorn group,Changshan-huyou Hawthorn group and Hawthorn Cassia seed could significantly remit liver weight increase.Spleen coefficient and thymus index results reveal that it is hard to determine whether the immune function of hyperlipidemia rat is affected.②Serum biochemical parameters reults showed that orally Changshan-huyou Hawthorn group could significantly improve the TC index.Gavaging Changshan-huyou Cassia seed group could significantly improve TG and LDL-C index,Gavaging Changshan-huyou Hawthorn group could significantly increase the HDL-C index.③ Liver sections showed that the model group has a higher degree of liver steatosis while Changshan-huyou Hawthorn group and Changshan-huyou Cassia seed group are much better.(4)The antioxidant experiments show that Changshan-huyou Hawthorn group and Changshan-huyou Cassia seed group could significantly improve SOD meanwhile lowering MDA.Changshan-huyou Hawthorn group,Changshan-huyou Cassia seed group and hawthorn Cassia seed group could improve GSH-PX apparently.Conclusion:Changshan-huyou is better than hawthorn and cassia seed in lipid dropping.Changshan-huyou Hawthorn group and Changshan-huyou Cassia seed formula show a good therapeutic effect on remitting blood lipid levels of hyperlipidemia.This study investigates themedical mechanism of Changshan-huyou and build a solid foundation for research and drug development. |
