| Rosa hybrida is a perennial woody plant that is widely used in gardens.In recent years,the chinese market has an increasing demand for new cultivars of Rosa hybrida,so there is a need for high-quality new cultivars.By summarizing the progress of plant breeding,it was found that chemical-induced polyploidy is a reliable method to obtain high-quality new cultivars.The study of chemical-induced polyploidy has been studied in many woody plants,but there are gaps in breeding studies of Rosa hybrida.Therefore,we chose to use chemical reagents to induce polyploid of Rosa hybrida.and successfully established a chemical-induced Rosa hybrida doubling system through a series of experiments,obtain polyploid plants at the same time.The results are as follows:1.The best proliferation medium for 4 monthly seasons(’Sunset Babylon Eyes’,’Red Apple’,’Pink Peerless Beauty’,’Micromoon’)was successfully screened.The experiment was set up with 4 treatments,each treated with 15 buds,3 replicates;One month later,the proliferation coefficient and bushbud height were counted and analyzed,and it was found that treatment 1(MS + agar(7.5 g/L)+ sucrose(30 g/L)+ NAA(0.1 mg/L)+ 6-BA(0.5mg/L))was the best treatment for the proliferation of ’Sunset Babylon Eye’,the proliferation coefficient of clump buds was about 3.42,the height of the clump buds was about 1.29 cm,and the bush buds grew well.Treatment 4(MS+agar(7.5 g/L)+ sucrose(30 g/L)+ NAA(0.1 mg/L)+ 6-BA(1.0 mg/L))was the best treatment for the proliferation of three kinds of bush buds(’Micromoon’,’Red Apple’ and ’Pink Peerless Beauty’),the proliferation coefficient of ’Micromoon’ was about 5.27,and the height of the clump buds was about 1.20 cm.The proliferation coefficient of ’Red Apple’ was about3.56,and the height of the clump buds was about 1.14 cm.The proliferation coefficient of’Pink Peerless Beauty’ was about 3.51,the height of the clump buds was about 1.07 cm.2.Using three reagents(colchicine,sulfamine and methalin)to induce ’sunset Babylon eyes’,all of which successfully obtained octaploid plant ’R8’.Colchicine has 3treatments,Oryzalin has 3 treatments,Pendimethalin has 3 treatments.Each treatment was 30 buds,3 replicates.A total of 24 octploid strains of ’Sunset Babylonian Eye’ were successfully induced by 12 treatments of colchicine,and considering the survival rate and induction rate,4 g/L+72 h was determined to be the optimal treatment for colchicine-induced doubling of ’Sunset Babylon Eyes’.A total of three octploid strains of’Sunset Babylonian Eye’ were successfully induced by the three treatments of Amisulf,and the survival rate and induction rate were comprehensively considered,and 60mg/L+24 h was determined to be the optimal treatment for the doubling of the ’Sunset Babylon Eye’ induced by Sulfamine.A total of 1 octuploidal strain of ’Sunset Babylonian Eye’ was successfully induced by three treatments of methalin,and considering the survival rate and induction rate,100 mg/L+24 h was determined to be the best treatment for colchicine-induced doubling of ’Sunset Babylon Eye’.3.Four Rosa hybrida(’Sunset Babylonian Eyes’,’Red Apple’,’Pink Peerless Beauty’,’Micromoon’)were induced by amsulin and methalin(’Sunset Babylon Eyes’,’Red Apple’,’Pink Peerless Beauty’,’Micromoon’),and three concentrations(0 mg/L,100 mg/L,200mg/L)were treated for 24 h,each treated with 30 lateral buds,and 3 replicates.In addition to successfully inducing ’Sunset Babylon Eyes’,only methalin induced ’Red Apple’ to obtain 4 octoid plants ’H8’,considering the survival rate and induction rate,100mg/L+24 h was the best treatment for doubling the inducing ’Red Apple’.The other two varieties failed to induce homozygous polyploidy,indicating that there was a great difference between different varieties.4.The propagation system of ’Sunset Babylonian Eyes’ octaploid ’R8’ was successfully established,and the ’R8’ proliferation medium was ’Sunset Babylonian Eyes’ tetraploid monthly medium(MS+agar(7.5 g/L)+ sucrose(30 g/L)+ NAA(0.1 mg/L)+6-BA(0.5 mg/L)),’R8’ increased to 3.30,and the height of seedlings was about 0.20 cm.The best bud medium for ’R8’ was screened,4 treatments were set up,5 buds per treatment,3 replicates,and the treatment 3(MS+agar(7.5 g/L)+ sucrose(30 g/L)+ NAA(0.1 mg/L)+ 6-BA(0.5 mg/L)+ GA3(0.2 mg/L))was determined as the best bud medium for ’R8’,the proliferation coefficient of ’R8’ was about 3.80,and the seedling height was about 1.19 cm.5.The phenotypic and physiological indexes of ’Sunset Babylonian Eye’ and ’Red Apple’ and their octuploids ’R8’ and ’H8’ were measured and compared,and it was found that octaploid plants had obvious growth retardation,but octoid plants had obvious stress resistance advantages.Both the ’R8’ and ’H8’ phenotypes showed slow growth rate,brittle plants with high water content and easy to break,few roots and short roots.Tetraploid under the same growth conditions,normal growth rate and developed root system.Compared with ’Sunset Babylon Eye’ and ’Red Apple’,the MDA content of ’R8’ and ’H8’decreased significantly,the POD content increased significantly,and the stress tolerance of polyploidy increased overall. |
