Local chicken breeds are one of the main sources of chicken meat in China,and lipid metabolism plays an important role in the growth of local chicken breeds.The molecular mechanism of lipid metabolism of No.2 Chahua broiler chickens with Yunnan characteristics was studied,and the scientific feeding cycle of No.2 Chahua broiler chickens was analyzed to provide a theoretical basis for improving the growth and reproduction,nutritional value and feeding methods of Yunnan local chicken breeds.In this study,140day-old male No.2Chahua broiler chickens were used as the research object.Thin layer chromatography(TLC)and real-time fluorescence quantitative polymerase chain reaction(q PCR)were used to study the transcriptional regulation mechanism of lipid metabolism and the expression of genes related to lipid metabolism in liver,pectoralis major and quadriceps femoris.1.The absorption and synthesis of TG in liver and skeletal muscle increased 16 h after feeding,suggesting that the intake of lipids before withdrawal was not enough to support the nutrient accumulation of skeletal muscle at 16 h after feeding,which was consistent with the decrease of TG and cholesterol ester levels in quadriceps femoris at this time.Therefore,the second feeding time of No.2 Chahua broiler chickens should be 16 h ago to maintain the normal growth and nutrient accumulation of No.2 Chahua broiler chickens.2.At 0.5h and 8h after feeding,the expression of lipid metabolism regulatory genes in liver and pectoralis major muscle of No.2 Chahua broiler chickens had no significant change,and the levels of TG and cholesteryl ester in liver and skeletal muscle had no significant change.Therefore,the energy intake of No.2 Chahua broiler chickens at 8h after feeding could maintain lipid homeostasis,and the time for the second feeding should be 8h later,in order to improve the effective utilization rate of feed and reduce feed waste.3.At 24 h after feeding,lipid decomposition,fatty acid transport and fatty acid oxidation metabolism in liver and skeletal muscle were active,gluconeogenesis in liver was enhanced,and the levels of TG and cholesterol ester in serum and skeletal muscle decreased.Therefore,the body needs to consume a large amount of stored nutrients to produce fatty acids to provide energy for the body.4.The liver and skeletal muscle responded adaptively to the deficiency of nutrients after fasting 16 h after feeding,suggesting that the time of the second regular feeding of No.2Chahua broiler chickens should be before 16 h,and after 8h when the nutritional level and lipid metabolism characteristics remained stable.Therefore,twice-daily feeding accords with the characteristics of lipid metabolism of No.2 Chahua broiler chickens and does not affect the normal nutrition accumulation,growth and development of No.2 Chahua broiler chickens. |
