| With the development of large-scale and intensive animal husbandry,the incidence of fatty liver disease in poultry has gradually increased.Animal fatty liver is a nutritional syndrome that usually occurs in poultry such as laying hens and laying ducks.Fatty liver can reduce feed utilization and production performance,and can seriously affect the health of poultry.Natural Chinese herbal medicine has a long history of use in China.It has the advantages of wide sources,low cost and little residue,so it has the potential to be used as feed additives.Increasing evidence suggests that adding specific herbal extracts to poultry diets can prevent and alleviate fatty liver.Uncaria,a commonly used traditional Chinese medicinal plant in Guangxi,is often used to treat hypertension and central nervous system diseases.Rhynchophylline,the main bioactive alkaloid component of Uncaria,has received attention in recent years for its blood pressure-lowering and neuroprotective effects.However,its role in regulating fatty liver has not been reported.In this study,we first conducted in vitro experiments by adding rhynchophylline to lipid accumulation models induced by oleic acid and palmitic acid in Hep G2(human liver cancer cells),LMH(chicken liver cancer cells),and ALM12(mouse liver cells),to observe its effect on lipid accumulation.Subsequently,we performed in vivo validation in C57BL/6J mice.The mice were divided into three groups: normal diet group,high-fat diet group,and high-fat diet group with rhynchophylline(40 mg/kg).The effects of rhynchophylline on lipid metabolism were verified through in vitro and in vivo experiments.Finally,we used molecular docking technology to find a possible target for the action of rhynchophylline——hepatic lipase,and conducted experimental validation.The research findings demonstrate that rhynchophylline effectively reduces lipid accumulation in human,chicken,and mouse liver cells,indicating its role in alleviating lipid metabolism disorders across different species.Furthermore,in vitro cell experiments show that rhynchophylline enhances liver cell energy metabolism,reduces oxidative stress,and improves glucose metabolism.In live mouse experiments,it was discovered that rhynchophylline lowers hepatic steatosis induced by a high-fat diet.Additionally,supplementation of rhynchophylline increases energy metabolism,improves glucose metabolism,and exhibits antioxidant effects in high-fat diet mice.To investigate the molecular mechanism by which rhynchophylline alleviates lipid metabolism disorders,this study utilized Discovery Studio software to perform molecular docking of rhynchophylline with lipid metabolism-related proteins in the Mala Cards database.It was found that rhynchophylline exhibited favorable interactions with hepatic lipase.Finally,experimental validation revealed that rhynchophylline enhances lipase activity,promoting fat decomposition and thus reducing lipid accumulation.This study,through in vivo and in vitro experiments,reveals that rhynchophylline alleviates lipid accumulation caused by a high-fat diet and helps prevent hepatic steatosis and fatty liver.It is noteworthy that rhynchophylline is widely available and cost-effective,providing new possibilities for the prevention and treatment of fatty liver in livestock and poultry. |
