Sow endometritis is a disease of reproductive disorders in sows.Its main clinical features are pus adherent to the tail,decreased childbirth,abnormal estrus,miscarriage,dystocia,and reproductive disorders in sows.The incidence of sow endometritis is high and it is difficult to cure.Once the sow becomes ill,the elimination rate is high,which leads to the elimination of too many young gilts in the breeding farm,which brings greater economic losses to the breeding farm.At present,the main method to treat the disease is the combined use of antibiotics with Chinese herbal medicines,vaccines,hormones,etc.Because antibiotics are the main drugs,all of them have the disadvantage of antibiotic resistance.Therefore,the discovery or development of new drugs or methods for the treatment of sow endometritis has important production significance.Recently,nanomaterials as a new type of materialhave been widely used in medical research,drug development,and environmental disinfection.Most nano-materials have strong antibacterial properties,which are pollution-free and don’t contribute bacterial resistance.Among them,nano-silver has good antibacterial properties and high biological safety,and is widely used in material medicine,drug carriers,environmental disinfection,and so on.Therefore,in this study,a safe and effective nano-silver solution was selected to wash the reproductive tract of sows to reduce pathogenic bacteria in the reproductive tract of sows and the inflammation of sow endometritis due to bacterial growth.This research aims to develop drug-free,high-efficiency and low-toxicity uterine infusions to remove pathogenic bacteria in the postpartum environment of sows.First,We cooperated with new material company to produce nano-silver nano-silver with uniform size,and physically characterized the material by scanning electron microscopy and Raman spectroscopy;secondly,it studied the effect of nano-silver aqueous solution on the most common pathogenic bacteria in the reproductive tract of sows through antibacterial experiments in vitro.The antibacterial properties of nano-silver to Staphylococcus aureus,Escherichia coli and Streptococcus were explored and the minimum inhibitory concentration was determined;finally,the safety evaluation test of the nano-silver at the cell level and the clinical efficacy evaluation test of sows with endometritis were carried out.The test results show that: 1)The nano-silver with high quality were homemade and SEM shows the nano-silver has small size and homogeneous dispersing.2)Nano silver has a certain bactericidal effect on standard bacteria and pathogenic bacteria related to sow endometritis(Gram-positive: Staphylococcus aureus and Streptococcus;Gram-negative bacteria:Escherichia coli),demonstrating that Nano silver can be used as a broad-spectrum bactericide.3)The minimum inhibitory concentration(MIC)of nano-silver used in this study was measured to be 62.5 μg/m L.After treated with nano-silver,the morphology and viability of HEK293 T cell were normal,indicating that nano-silver is non-toxic to cells cultured in vitro.Toxicity experiments in mice also showed that the uterine morphology of the female mouse after nano-silver perfusion did not have any obvious abnormalities,which further showed that the nano-silver solution within a certain concentration range is safe to use as a uterine perfusion agent;4)In clinical trials,compared with the control group without treatment,the pus phenomenon of sows with endometritis after nano-silver perfusion was significantly reduced,and the detection of physiological and biochemical indicators also found that the number of white blood cells was reduced,indicating that nano-silver perfusion can alleviate endometritis related symptoms.The experimental results of this study show that nano-silver as a new green,harmless and broad-spectrum antibacterial and non-drug resistant perfusion agent can effectively treat sow endometritis.
