Objective : The comprehensive evaluation of the antibacterial effect of Cyperus esculentus on Staphylococcus aureus and Escherichia coli can provide theoretical support and technical reference for the rational utilization of Cyperus esculentus resources.As a natural plant antibiotic,it has very important production and practical significance for the utilization of Cyperus esculentus.Methods :(1)In the experiment,different parts of Cyperus esculentus extract : Cyperus esculentus tuber,Cyperus esculentus stem and leaf,Cyperus esculentus tuber and stem and leaf were mixed in equal proportions into three treatments,and the concentration was set to four gradients : 50μL,70μL,150μL,250μL.The bacteriostatic test(Oxford cup method,morphological observation of bacteria),the growth curve(adding the best part and content of bacteriostatic extract to the bacterial solution),and the analysis of metabolism(metabolome).(2)Taking Cyperus esculentus as the research object,the metabolites of Cyperus esculentus were analyzed,and the main antibacterial components in Cyperus esculentus were further screened out,and the main components were quantitatively analyzed by liquid chromatography.(3)Antibacterial test : K-B method and morphological observation of bacteria.The k-B method used the main components of Cyperus esculentus extract,naringenin,diosmetin,acacetin,and silybinin A as four treatments,and each treatment set four concentrations.Growth law : Response surface analysis combined with experiment 1 was used for experimental design.Finally,four components were added to the two bacterial solutions for metabolic analysis.Results :(1)In the experiment,Cyperus esculentus extract had antibacterial effect on Escherichia coli and Staphylococcus aureus.The higher the content of Cyperus esculentus extract(four gradients),the better the antibacterial effect;scanning electron microscopy showed that the extracts of different parts of Cyperus esculentus had obvious damage to the two bacteria;the growth curve showed that the antibacterial effect was more significant with the increase of time.The metabolites such as levofloxacin,tunicamycin A,chlorogenic acid and fluoroerythromycin in Cyperus esculentus have antibacterial effects.(2)In this part of the experiment,the four substances of naringenin,diosmetin,acacetin and silybinin A in the metabolites(flavonoids)of Cyperus esculentus were determined,and the content of the four substances in the extract of Cyperus esculentus stems and leaves was determined.(3)In this experiment,naringenin,acacetin,diosmetin and silybinin A had good antibacterial effect on Escherichia coli and Staphylococcus aureus.Each substance alone or in combination has a good effect on inhibition.Scanning electron microscopy was used to observe the damage of four substances to bacteria.A variety of amino acids,lincomycin,sulfadiazine and doxycycline of metabolites in the five experimental groups played an antibacterial role.Conclusion : Cyperus esculentus has antibacterial effect on Escherichia coli and Staphylococcus aureus.The extract of Cyperus esculentus mixture had the best antibacterial effect,and the longer the inhibition time of Cyperus esculentus on bacteria,the better the antibacterial effect(within 24h).The content of flavonoids in Cyperus esculentus in Cyperus esculentus extract was as follows : naringenin 8.14423μg /m L,diosmetin 5.58963μg / m L,acacetin 4.18378μg / m L,silybinin A3.67256μg / m L.The four substances have good antibacterial effects on both bacteria.The same metabolites in the experimental group,multiple amino acids,lincomycin,sulfadiazine and doxycycline play a role in antibacterial. |
