| Agropyron cristatum is a Gramineae plant,which belongs to the Gaertn.,is a perennial herb.It has a wide range of distribution,especially in Inner Mongolia,China,which has the highest density and strong resistance to stress.A.cristatum has high feeding value,ecological value and germplasm resource value.This study used SSR molecular labelling technology to evaluate the genetic diversity of 10 wild germplasm resource in central Inner Mongolia,and determined the differences between different germplasm resources by analysing phenotypic trait data.The results are as follows:1.There are significant genetic differences in the 27 main phenotypic traits of A.cristatum germplasm.The six personality variation coefficients of mechanical organization of leaf,leaf sheath cuticle of upper epidermal,leafcuticle of lower epidermal,epidermal cells on leaf sheaths,bulliform cells,and seed number all exceed 30%,which can be used as an indicator of resource.The genetic diversity index of the quantitative characteristics of A.cristatum germplasm resources for testing varies from 6.94 to 7.63 between groups.2.A total of 106 alleles were detected by screening 12 primer,with an average of 8,Ne with an average of 4.59,Ho with an average of 0.41,He with an average of 0.68,UHe with an average of 0.71,and I with an average of 1.54,indicating a high genetic diversity among 10 materials.3.A.cristatum group showed a low degree of differentiation,with a gene flow of3.45~29.27,indicating frequent gene exchange between icegrass groups.Molecular variance analysis shows that 62.4% of the genetic differences in the A.cristatum population come from within the population,and 37.6% of the genetic differences come from between groups.SSR cluster trees can be divided into four categories with a genetic coefficient of about 0.49.When the classification number K is 3,the value of ΔK(delta K)reaches the maximum,indicating that the differentiation of the group is the most obvious.The whole group of A.cristatum is divided into three subgroups,and there are obvious differences between them.The existence of genes may be caused by frequent gene flow or inbreeding between these individuals.4.The association analysis of A.cristatum phenotypic traits and SSR molecular markers detected that 6 personality traits are associated with 10 SSR markers,and the germination rate is significantly related to 2 sites,indicating that the trait is jointly regulated by multiple genes;J-29 marker are significantly related to multiple traits,reflecting gene pleiotropy. |
