Cashmere goats are very characteristic biological resource used for both cashmere and meat in China.The cashmere produced by cashmere goat is famous all over the world for its soft texture and good thermal insulation,and has high economic value.Cashmere is the product of secondary hair follicle development,which go through three stages: telogen,anagen and catagen with hair follicle growth.In the three stages of hair follicle growth,proteins,as the executor of life activities,play an important role.In this study,Albas white cashmere goats taken as research objects,proteome sequencing data of hair follicles at different growth stages was analyzed to screen the key proteins.Meantime,the interaction protein,physical and chemical properties and protein structure were predicted,tissue expression experiment and a protein database construction were instigate.The results are as follows:(1)FKBP10 and FBN2 were selected as the key proteins in the cycle growth of cashmere goat hair follicles,and ALB,COL1A1 and ELN have been predicted as their interaction proteins.(2)A Database of proteins in cashmere goat hair follicle growth has been established.(3)FKBP10,FBN2,COL1A1 and ALB have slightly hydrophobic,ELN has slightly hydrophilic,and they are all located on the outside of the membrane,FKBP10,FBN2 and COL1A1 are secretory proteins that can bind to the receptor proteins on the membrane.(4)The results of q PCR,Western-blot and immunohistochemical showed that there are significant differences in the expression of FKBP10,FBN2,COL1A1,ELN,ALB and their genes at different hair follicle growth stages.The expression is higher in anagen and lower in telogen,and all of them are highly expressed in the outer root sheath of secondary hair follicles in anagen,which can promote hair follicle growth.The results of this study provide a theoretical basis for revealing the growth mechanism of hair follicles and villi in cashmere goats. |
