Oregano(Origanum vulgure)is a perennial aromatic plant of the Labiaceae.Essential oil of oregano is an effective anti-microbial substance that can be used as the substitute of antibiotics in animal feed.This thesis aimed to further modify rapid propagation,seedling culture and transplantation techniques to provide technical solutions for large-scale cultivation and production of oregano.The results are as follows.(1)The in vitro proprogation system of oregano was improved by screening suitable basal salt mediums,plant growth regulators and concentrations of activated charchoal.The experimental results showed that 1×MS+2.4 mg·L-1 GA+2.0 mg·L-1 6-BA+0.1 mg·L-1 NAA or 1×MS+2.0 mg·L-16-BA+0.1 mg·L-1NAA were both suitable for seed germination of oregano;and 0.5 to 0.75×MS basal medium was suitable for the growth of oregano with the highest number of branches,root length,plant height and fresh weight per plant.Different combinations and concentrations of four different growth regulators(IBA,NAA,6-BA and KT)were screened for their effects on oregano’s survival rate,rooting rate,the number of branches per plant,root length,plant height and whole plant fresh weight.Result based on comprehensive evaluation showed that 2.0 mg·L-1 NAA+0.25 mg·L-1 6-BA or 1.0 mg·L-1 NAA both promoted the growth of the aboveground biomass but no the roots,which thereof can be used for the proliferation culture of oregano.Supplementation of 0.1 mg·L-1 NAA,0.4 mg·L-1 IBA,0.8 mg·L-1 IBA+0.25 mg·L-1 IBA,or 0.8 mg·L-1 IBA promoted the growth of the aboveground and root development of oregano.In addition,supplementation of 1500mg·L-1 AC effectively improved the plant height,the number of branches,root length,and fresh weight.(2)The suitable culture medium and nutrient solution used in oregano cutting propagation were improved.The results showed that the fresh weight,branches,plant height,total root length,root surface area,root volume,root tip numbers,capillary root length and total volume of oregano cuttings increased first and then decreased with the increase of the concentration of macroelements in nutrient solution.The effect of0.25×modified Hoagland’s nutrient solution was the best.Compared with the control,fresh weight and root volume of plants increased by 242.47%and 463.6%,respectively.When peat soil and vermiculite with different volume ratio were used as cutting medium and 0.25×nutrient solution was irrigated,the survival rate and growth indexes of oregano cuttings also showed significant differences.Among them,the survival rate of Greek oregano cuttings was 100%in the peat soil vermiculite volume ratio of 25:75,and the cuttings growth and root development were the best.Therefore,using peat soil:vermiculite(25:75)as cutting medium and watering 0.25×nutrient solution every week is the suitable method for propagation of oregano clones.(3)The weed control after oregano seeding transplantation in field were studied.The results of field experiments showed that pre-emergent herbicides and Plastic mulchs had significant effects on weed control and oregano growth.Among them,black Plastic mulchs and spraying 2625 m L hm-2pendimethalin 33%EC or 1200 m L·hm-2 S-metolachlor EC 960 g·L-1 had the best control effect on weeds,and the control effect in spring was 93.60%~97.60%.However the pot experiment showed that acetochlor EC,pendimethalin EC and S-metolachlor EC had certain inhibitory effect on growth and development of oregano.Spraying 2250 m L·hm-2 pendimethalin 33%EC had no significant inhibitory effect on the number of branches per plant,the number of buds,plant height,aboveground and underground fresh weight,root length,stem leaf ratio and photosynthetic rate of oregano.In addition,in the summer of high temperature and humidity,the Plastic mulchs should be removed in time to improve the soil permeability,so as to promote the secondary growth of oregano.
